Towards detection of SARS-CoV-2 RNA in human saliva: A paper-based cell-free toehold switch biosensor with a visual bioluminescent output

Summary: The COVID-19 pandemic has highlighted the global demand for rapid, low-cost, widely distributable and point-of care nucleic acid diagnostic technologies. This study developed a paper-based nucleic acid diagnostic platform using lyophilized cell-free protein synthesis and toehold switch riboregulators, which can be activated simply by the addition of saliva, at a low estimated cost.

Summary: Bioavailable glutamine (Gln) is essential for metabolism, intestinal health, immune function, and cell signaling. Routine measurement of serum Gln concentrations can aid in the improved diagnosis and treatment of severe infections, anorexia nervosa, chronic kidney disease, diabetes, and cancer, yet current methods may underutilize patient Gln levels. The use of Cell-free protein synthesis (CFPS) as a sensing platform for detecting Gln concentrations in human samples shows promising clinical applications and may lead to transformative point-of-care diagnostics and treatment regimens for various diseases.

Evaluation of an E. coli Cell Extract Prepared by Lysozyme-Assisted Sonication via Gene Expression, Phage Assembly and Proteomics

Summary: This study evaluated the use of E. coli cell extract prepared with lysozyme incubation and sonication, demonstrating successful cell-free expression of YFP and T7 bacteriophages. Quantitative proteomics was used to compare protein composition between extracts, showing minimal differences in LAS extracts but increased release of DNA-binding proteins with more sonication cycles.

Summary: By measuring dynamic mRNA and protein levels in genetic construct libraries, the effects of cosolutes on individual gene expression processes were studied. Cosolutes were found to have different effects on transcription and translation rates, with trade-offs between time delays, expression tunability, and maximum productivity. Biophysical models can predict translation rates in TX-TL, showcasing the potential for optimizing performance in various cell-free applications.
