Bacillomycin D-C16 inhibits growth of Fusarium verticillioides and production of fumonisin B1 in maize kernels

Fuxing Lin et al.

Summary: This study investigated the inhibitory effects of Bacillomycin D-C16, a natural antimicrobial lipopeptide produced by Bacillus subtilis, on the growth and toxin production of Fusarium verticillioides in maize. The results showed that Bacillomycin D-C16 displayed strong fungicidal activity against F. verticillioides and significantly reduced the production of fumonisin B-1.

Exploring the Molecular Basis of Substrate and Product Selectivities of Nocardicin Bifunctional Thioesterase

Qian Yu et al.

Summary: The study showed that NocTE controls substrate selectivity through factors such as peptide length and the β-lactam ring, capturing key residues involved in this process. Furthermore, it was found that natural product liberation from the active pocket differed between the product undergoing epimerization and its diastereomer, explaining their biological activity differences.

CodY, ComA, DegU and Spo0A controlling lipopeptides biosynthesis in Bacillus amyloliquefaciens fmbJ

J. Sun et al.

Summary: This study investigated the regulatory effects of genes codY, comA, degU, and spo0A on the biosynthesis of three lipopeptides in Bacillus amyloliquefaciens. Deletion and overexpression of these genes significantly impacted the production of lipopeptides, with spo0A showing potential as a key regulator for bacillomycin D synthesis. These findings suggest genetic engineering of regulatory genes as an effective strategy for improving antimicrobial lipopeptide yields in industrial production.

Biosynthetic Cyclization Catalysts for the Assembly of Peptide and Polyketide Natural Products

Maria L. Adrover-Castellano et al.

Summary: This review explores the general cyclization mechanism of TE domain in NRPS and PKS, including oligomerization and CLCs. It also discusses other cyclization catalysts such as C-T domains, R domains, R-D domains, Dieckmann cyclases, among others. The diversity of cyclization catalysts highlighted in the studies for the production of biologically relevant, complex cyclic natural products and related compounds is summarized here.

The inherent flexibility of type I non-ribosomal peptide synthetase multienzymes drives their catalytic activities

Sarah Bonhomme et al.

Summary: NRPSs are multienzymes that produce complex natural metabolites with applications in medicine and agriculture. They consist of multiple catalytic domains and non-catalytic carrier protein domains, allowing flexibility in catalyzing and transporting products. Recent studies providing structural views of multi-domain NRPSs have enhanced understanding of their catalytic cycle.
