DNAzyme-based colorimetric assay and its application for lipopolysaccharide analysis assisted by oxime chemistry

Summary: This study proposed for the first time that the cleavage activity of DNAzyme is associated with the release of hydroxyl ions, which can be utilized for colorimetric assay. Using this property, a colorimetric strategy for lipopolysaccharide (LPS) analysis was constructed. By linking DNAzyme fragments modified with aldehyde and hydroxylamine groups through oxime chemistry, and with LPS preventing the formation of oxime bond, a successful DNAzyme-based colorimetric assay was developed.

The performance of this biosensor in different cell lysates demonstrated great potential for assay of microRNAs at low expression levels in complex samples.
