Hallmarks of Cancer: New Dimensions

Douglas Hanahan

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Dynamic Monitoring of EMT in CTCs as an Indicator of Cancer Metastasis

Zhen Zhang et al.

Summary: The study developed a multiplex surface-enhanced Raman scattering nanotechnology to characterize EMT-associated phenotypes in CTCs, observing changes in markers during EMT process in breast cancer models and successfully detecting disease in patient samples. This approach provides insights into monitoring cancer metastasis and potential for early detection.

Simultaneous biochemical and functional phenotyping of single circulating tumor cells using ultrahigh throughput and recovery microfluidic devices

Yang Liu et al.

Summary: The combinational approach of using i(2)FCS method enables efficient processing of whole blood samples for profiling patient-derived CTCs, providing adequate and functional CTCs for subsequent studies. This method allows for the query of CTCs with single-cell resolution based on their expression of phenotypic surface markers and migration properties, revealing dynamic phenotypes and high-motility subpopulation of CTCs in blood samples from metastatic lung cancer patients.
