Intrinsically disordered proteins play diverse roles in cell signaling

Sarah E. Bondos et al.

Summary: Signaling pathways allow cells to detect and respond to various chemical and physical stimuli, forming a communication network that regulates cell activities and coordinates cell function. Successful signaling requires proteins that can form active and inactive states and engage in multiple interactions. Intrinsically disordered proteins play a critical role in cell signaling, present in diverse organisms and stages of signaling pathways.

Mechanical instability of adherens junctions overrides intrinsic quiescence of hair follicle stem cells

Ritusree Biswas et al.

Summary: Vinculin, a key mechanotransducer in cell junctions, is essential for maintaining the quiescence of bulge stem cells and regulating hair follicle cycling. Conditional knockout of vinculin in murine skin leads to weakened AJs, despite increased expression of Ecadherin and α-catenin. Mechanistically, vinculin functions by maintaining α-catenin in a stretched/open conformation, which controls the retention of YAP1 at the AJs, revealing a novel regulatory link between mechanical stability of cell junctions and maintenance of stem cell quiescence.

Activated nanoscale actin-binding domain motion in the catenin-cadherin complex revealed by neutron spin echo spectroscopy

Bela Farago et al.

Summary: The cadherin-catenin complex transduces mechanical tension in cell-cell adhesion, with the actin-binding domain adopting various configurations. Using neutron spin echo spectroscopy and theoretical physics analyses, the protein dynamics of the complex were determined. The motion of the entire actin-binding domain becomes activated on nanosecond to submicrosecond timescales, showing the formation of an entropic trap that impedes binding to actin under zero force.

Sorting of cadherin-catenin-associated proteins into individual clusters

Regina B. Troyanovsky et al.

Summary: The study explores the structure and function of the multiprotein complex formed by the cytoplasmic tails of classical cadherins in adherens junctions. It reveals that cytoplasmic proteins interact with this complex and organize into clusters, potentially synchronizing signaling networks among neighboring cells within tissues.

Distinct intramolecular interactions regulate autoinhibition of vinculin binding in αT-catenin and αE-catenin

Jonathon A. Heier et al.

Summary: The study reveals that alpha T-catenin has similar binding characteristics to alpha E-catenin but with differences in affinity and conformation when binding to vinculin. The N-terminus of alpha T-catenin is crucial in maintaining M-region autoinhibition and recruiting vinculin to cell-cell contacts.
