Structure-based design, synthesis and antiproliferative action of new quinazoline-4-one/chalcone hybrids as EGFR inhibitors

Summary: A new series of quinazoline-4-one/chalcone hybrids, 7-26, were synthesized and tested as EGFR inhibitors with antiproliferative activity. Three compounds showed the greatest antiproliferative activity and were the most potent EGFR inhibitors. These three compounds improved the levels of active caspase-3, 8, and 9, induced cytochrome c and Bax levels, and down regulated Bcl_2 levels. The most active inhibitors docked well inside EGFR active sites.

Mechanistic study of the antibacterial potential of the prenylated flavonoid auriculasin against Escherichia coli

Summary: Bacterial resistance is a growing concern and the development of new antibacterial agents is not keeping pace. Prenylated flavonoids have shown promise as a class of natural antibiotics that can combat resistant pathogens. In this study, a large library of flavonoids was screened for their potential to inhibit the B-subunit of gyrase, a bacterial enzyme. Several prenylated flavonoids showed promising results, with auriculasin being the most potent antibacterial candidate. Mechanistic analysis revealed that auriculasin inhibits the Gyr-B enzyme and further experiments provided insights into its mode of action. This research highlights the potential of prenylated flavonoids as antibacterial agents and provides valuable information for the development of more potent derivatives.

Discovery of a potent β-catenin destabilizer for overcoming the resistance of 5-fluorouracil in colorectal cancer

Summary: This study synthesized novel stemona alkaloid analogues, among which compound 3 was found to efficiently inhibit various colorectal cancer cells, including drug-resistant cells, by reducing the protein level of beta-catenin without affecting its mRNA level. The downstream proteins expression was also significantly inhibited, suggesting potential for compound 3 as a destabilizer of beta-catenin for the treatment of colorectal cancer patients.

New chalcone derivatives as potential antimicrobial and antioxidant agent

The compounds exhibited moderate to good activities against Staphylococcus aureus, Bacillus subtilis, Salmonella typhi, Escherichia coli, and Aspergillus niger, with compound 6c showing the strongest antioxidant activity. Absorption, distribution, metabolism, and excretion properties of the compounds were evaluated using SwissADME, with compound 6e identified as a lead-like molecule based on lead likeness assessments.

The presence of -COOH functionality in complex 2 led to higher cytotoxicity compared to complex 1. The complexes promoted cellular detachments via nuclear condensation and membrane destabilization followed by apoptosis as a result of metal-assisted generation of reactive oxygen species.

Discovery of novel JAK2 and EGFR inhibitors from a series of thiazole-based chalcone derivatives

Summary: In this study, aromatic alkyl-amino analogs of thiazole-based chalcone were investigated for their inhibitory activity against JAK2 and EGFR proteins. Compounds 11 and 12 showed susceptibility in HEL cells, while compound 25 was effective against A431 cells. Compound 25 was identified as a dual inhibitor against both JAK2 and EGFR, while compounds 11 and 12 selectively targeted JAK2. Through molecular docking analysis, the compounds were found to interact with specific residues at the catalytic sites of the proteins. These thiazole derivatives show potential as promising targeted drugs for cancer therapy.
