Pharmacokinetics and Pharmacodynamics of Clofazimine for Treatment of Cryptosporidiosis

Summary: Infection with Cryptosporidium spp. can cause severe diarrhea, and the existing drug for treatment, nitazoxanide, has limited efficacy. A previous clinical trial showed that the antimycobacterial therapeutic clofazimine had potential in treating Cryptosporidium infection, but its concentration in patients did not reach the required level for efficacy.

The Effect of Short-Chain Fatty Acids on Growth of Cryptosporidium parvum In Vitro

Summary: This study examined the effect of short-chain fatty acids (SCFAs) on the growth of Cryptosporidium in human cells and found that acetate, propionate, and butyrate have direct inhibitory activities against the parasite. Additionally, these SCFAs may inhibit infection by inducing apoptosis and inflammatory responses.

Clofazimine for Treatment of Cryptosporidiosis in Human Immunodeficiency Virus Infected Adults: An Experimental Medicine, Randomized, Double-blind, Placebo-controlled Phase 2a Trial

Summary: The study evaluated the efficacy, pharmacokinetics, and safety of clofazimine in HIV patients with cryptosporidiosis. Results showed that clofazimine did not significantly reduce Cryptosporidium shedding, but led to more adverse effects in severely immunocompromised HIV patients. This suggests limitations in the safety and efficacy of clofazimine for treating cryptosporidiosis in this patient population.
