Dually Active Polycation/miRNA Nanoparticles for the Treatment of Fibrosis in Alcohol-Associated Liver Disease

Chuhan Zhang et al.

Summary: This study developed dual-functioning nanoparticles for the effective delivery of antifibrotic RNA and CXCR4 inhibitor, which shows promise for improving the treatment of alcohol-associated liver disease. The nanoparticles were able to significantly silence miR-155 expression and reduce liver damage in a model.

Nanoparticle Delivery of Novel PDE4B Inhibitor for the Treatment of Alcoholic Liver Disease

Jingyi Ma et al.

Summary: This study investigates the potential use of a novel PDE4B inhibitor, KVA-D88, for the treatment of ALD. The inhibitor showed promising results in inhibiting inflammatory response and promoting liver cell health. Encapsulating KVA-D88 in nanoparticles improved its solubility and effectiveness in vivo. The study shows that KVA-D88-loaded NPs have the potential to effectively treat ALD in mice.

The Integrated Multiomics Landscape at Peak Injury and Resolution From Alcohol-Associated Liver Disease

Sukanta Das et al.

Summary: This study identified the liver transcriptome, fecal microbiome, and portal serum metabolome involved in the pathogenesis and resolution of alcohol-associated liver disease (ALD). Gender-specific differences were found in the changes during ALD resolution. The integration of multiomics data identified Map3k1 as a key gene driving ALD pathogenesis and resolution.

The Gut-Liver Axis in Chronic Liver Disease: A Macrophage Perspective

Kevin De Muynck et al.

Summary: Chronic liver disease is a growing health concern, commonly associated with obesity, alcohol overconsumption, and progressive cholestasis, leading to persistent low-grade inflammation and advancing fibrosis. The pathophysiology of CLD also involves intestinal dysbiosis, bile acid dysregulation, and gut barrier disruption, where macrophages play a crucial role in disease progression and intestinal barrier breakdown.

Oxy210, a novel inhibitor of hedgehog and TGF-β signalling, ameliorates hepatic fibrosis and hypercholesterolemia in mice

Simon T. Hui et al.

Summary: NASH is associated with increased morbidity and mortality in NAFLD patients, and liver fibrosis is a strong prognostic factor. Oxy210 reduces hepatic fibrosis in NASH by inhibiting Hh and TGF-beta signaling. In mouse studies, Oxy210 not only decreased liver fibrosis but also improved hypercholesterolemia.
