Enzymatic cascade reaction in simple-coacervates

Oscillating the local milieu of polymersome interiors via single input-regulated bilayer crosslinking and permeability tuning

Summary: The permselectivity of cellular membranes is crucial for maintaining intracellular homeostasis. However, it is challenging to synergize the stability and permeability of artificial cells and imitate local milieu fluctuations. In this study, a concurrent cross-linking and permeabilizing strategy was developed using pH-responsive polymersomes containing Schiff base moieties within bilayer membranes. The strategy allowed precise control of mesh sizes of the crosslinked bilayers, resulting in discriminative permeability to molecules of specific sizes. The permselectivity of the bilayer membranes enabled intravesicular pH oscillation and the dissipative self-assembly of pH-sensitive dipeptides. Additionally, the permeabilization of polymersomes could be regulated by intracellular pH gradient, allowing controlled release of encapsulated payloads.
