Rhynchophylline ameliorates cerebral ischemia by improving the synaptic plasticity in a middle cerebral artery occlusion induced stroke model

Lei Wang et al.

Summary: This study aimed to investigate the role of rhynchophylline in neural function and neuroplasticity in a rat model of cerebral ischemia. The results showed that rhynchophylline improved sensory-motor function, alleviated memory impairment, and enhanced synaptic plasticity. Rhynchophylline may have potential therapeutic effects in ischemic stroke.

Hyperexcitable arousal circuits drive sleep instability during aging

Shi-Bin Li et al.

Summary: Sleep quality declines with age and this study identified hyperexcitable hypocretin/orexin (Hcrt/OX) neurons as drivers of sleep fragmentation during aging. Activation of Hcrt neurons increased wakefulness and aged Hcrt neurons showed hyperexcitability and impaired M-current. Disruption of Kcnq2/3 genes in young mice led to sleep destabilization resembling aging-associated sleep fragmentation.

Single-cell transcriptomics and cell-specific proteomics reveals molecular signatures of sleep

Pawan K. Jha et al.

Summary: This study used single-cell RNA sequencing and cell-type-specific proteomics to investigate the molecular mechanisms of sleep. The findings suggest that different cell types in different brain regions exhibit diverse transcriptional responses to sleep need and sleep deprivation affects the expression of proteins and phosphorylation in astrocytes and neurons.

Rhynchophylline Administration Ameliorates Amyloid-β Pathology and Inflammation in an Alzheimer's Disease Transgenic Mouse Model

Wing-Yu Fu et al.

Summary: The study found that rhynchophylline can improve amyloid plaque burden and reduce inflammation in an Alzheimer's disease mouse model. Transcriptome analysis revealed that rhynchophylline regulates multiple molecular pathways associated with amyloid metabolism and inflammation, suggesting potential therapeutic benefits for Alzheimer's disease.

A spatially resolved brain region- and cell type-specific isoform atlas of the postnatal mouse brain

Anoushka Joglekar et al.

Summary: The study revealed variations in alternative RNA splicing across different brain regions, with unclear single-cell resolution of such differences. Certain genes can be influenced by multiple cell types, allowing regional identity to occasionally override cell-type specificity. Through spatial transcriptomics and long-read sequencing, a deeper understanding of brain integration of molecular and cellular complexities is achieved.

Altered hippocampal transcriptome dynamics following sleep deprivation

Marie E. Gaine et al.

Summary: Widespread sleep deprivation poses a significant public health issue globally, impacting both adolescents and adults. Acute sleep deprivation affects hippocampal gene expression, leading to changes in various biological processes. Recovery sleep following acute sleep deprivation partially normalizes gene expression levels in the hippocampus, demonstrating the complex and differential regulation of genes by sleep deprivation.

Cellular Effects of Rhynchophylline and Relevance to Sleep Regulation

Maria Neus Ballester Roig et al.

Summary: Uncaria rhynchophylla, a plant used in traditional Chinese and Japanese medicines, has shown to have beneficial effects on sleep quality and duration. The alkaloid component rhynchophylline in Uncaria species may modulate sleep through pathways involving ion channels, receptors, and protein kinases, ultimately impacting neuronal communication and sleep phenotypes. Understanding the mechanisms of action of rhynchophylline could have implications for sleep pharmacology.
