2,3,4′,5-Tetrahydroxystilbene-2-O-β-D-glucoside ameliorates gentamicin-induced ototoxicity by modulating autophagy via Sesn2/AMPK/mTOR signaling

Yu-Hsuan Wen et al.

Summary: This study investigated the otoprotective mechanism of 2,3,4',5-tetrahydroxystilbene-2-O-beta-D-glucoside (THSG) in attenuating gentamicin ototoxicity. THSG was found to decrease gentamicin-induced ototoxicity in UB/OC-2 cochlear cells via regulating the autophagic signaling involving Sesn2/AMPK/mTOR pathway.

Chloroquine may induce endothelial injury through lysosomal dysfunction and oxidative stress

PauloC Gregorio et al.

Summary: COVID-19 pandemic continues with limited antiviral agents available. Chloroquine, despite showing in vitro antiviral activity, had disappointing results in clinical trials and may harm outcomes. The adverse effects of chloroquine on endothelial cells were mitigated by agalsidase-beta treatment, suggesting a potential role of endothelial cell injury in the failure of chloroquine therapy for COVID-19.

A Novel Spo11 Homologue Functions as a Positive Regulator in Cyst Differentiation in Giardia lamblia

Yu-Chien Chen et al.

Summary: This study investigated the role of the topoisomerase Spo11 in the encystation process of Giardia lamblia. The findings indicate that Spo11 acts as a positive regulator in activating encystation, interacting with transcription factors involved in cwp gene expression for cyst formation. Disruption of the spo11 gene led to decreased cwp gene expression and cyst formation, highlighting the importance of Spo11 in regulating Giardia differentiation into cysts.

A Novel Multiprotein Bridging Factor 1-Like Protein Induces Cyst Wall Protein Gene Expression and Cyst Differentiation in Giardia lamblia

Shao-Wei Huang et al.

Summary: The study revealed that Giardia's MBF1 gene plays a crucial role in cyst differentiation by binding to key promoter regions of cwp1-3 and myb2 genes and interacting with transcription factors to up-regulate cwp gene expression during encystation. This positive regulation of MBF1 in Giardia suggests a functional conservation of the gene in driving cyst formation.

Giardia duodenalis: Biology and Pathogenesis

Rodney D. Adam

Summary: Giardia duodenalis, initially observed by Leeuwenhoek in 1681, gained attention as a human pathogen in the 1960s. Advances in technology have provided insights into its biology and immunology, revealing the importance of a balanced host immune response and the role of the Th17 response. Infection with Giardia can result in a wide range of symptoms, highlighting the complexity of host-parasite interactions.

The development of an endoplasmic reticulum-targeting fluorescent probe for the imaging of 1,4-dithiothreitol (DTT) in living cells

Abdul Hadi Mehmood et al.

Summary: 1,4-Dithiothreitol (DTT) is a crucial reducing agent involved in protein folding and ER homeostasis. Abnormally high levels of DTT can cause ER stress and cell death. A newly designed ER-targeting fluorescent probe, ER-DTT, based on naphthalimide, p-toluenesulfonamide, and sulfoxide, showed good selectivity and sensitivity for imaging DTT in living cells.
