Microglia and their LAG3 checkpoint underlie the antidepressant and neurogenesis-enhancing effects of electroconvulsive stimulation

Neta Rimmerman et al.

Summary: This study highlights the important role of microglia in the treatment of depression. Depletion and inhibition of microglia significantly affect the therapeutic effects of antidepressants, particularly in gene expression related to neurogenesis and dopamine signaling. The findings suggest that brain microglial LAG3 may be a promising target for novel antidepressant therapeutics.

A loss of mature microglial markers without immune activation in schizophrenia

Gijsje J. L. J. Snijders et al.

Summary: The density and morphology of microglial cells in schizophrenia patients were found to be unchanged, while the expression of several microglial-specific genes was decreased. This indicates a change in microglial phenotype rather than density, which could provide a crucial molecular hallmark for future research into the role of microglia in schizophrenia and other psychiatric disorders.

Complement-associated loss of CA2 inhibitory synapses in the demyelinated hippocampus impairs memory

Valeria Ramaglia et al.

Summary: The complement system is involved in synapse loss in the hippocampus of patients with multiple sclerosis, particularly in the CA2 subfield. This loss is associated with cognitive impairment, and in a mouse model of demyelination, C1q deposits were found in the CA2 pyramidal layer along with inhibitory synapses engulfed by microglia/macrophages, leading to impaired feedforward inhibition and memory deficits.

Regulation of microglia population dynamics throughout development, health, and disease

Brady P. Hammond et al.

Summary: The dynamic changes in microglia density, volume, and proliferation in the central nervous system play a crucial role in their function, with fluctuations affecting their ability to respond effectively to various stimuli. Microglia exhibit a robust proliferative capacity during development and in neurodegenerative diseases, but this capacity is largely absent at homeostasis. Proper microglial function in different states is heavily dependent on density fluctuations primarily induced by proliferation.

A lymphocyte-microglia-astrocyte axis in chronic active multiple sclerosis

Martina Absinta et al.

Summary: Through MRI-guided single-nucleus RNA sequencing, a diversity of glial and immune cells at the edge of demyelinated white matter lesions in multiple sclerosis (MS) have been identified, along with the definition of inflamed glial cell phenotypes in MS. The overlap of the transcriptional profile of microglia inflamed in MS with that in other neurodegenerative diseases suggests common mechanisms between primary and secondary neurodegeneration that could benefit from similar therapeutic approaches. The critical role of complement component 1q (C1q) in the activation of microglia inflamed in MS has been identified and potential therapeutic avenues, such as C1q inhibition, have been suggested for addressing chronic white matter inflammation.

Gene expression studies in Depression development and treatment: an overview of the underlying molecular mechanisms and biological processes to identify biomarkers

Nicole Mariani et al.

Summary: The development of major depressive disorder (MDD) is thought to be influenced by a combination of genetic, environmental, and psychological factors, as well as immune system, stress response, and neurotransmitter regulation, leading to a focus on the interplay between the periphery and the Central Nervous System (CNS). Studies have analyzed gene expression levels in postmortem brain tissue and blood samples of MDD patients, with a particular interest in mRNA levels as biomarkers for predicting therapy outcomes. Additionally, the choice of antidepressant medication for each individual still remains largely uncertain.
