Deciphering the mechanism of p73 recognition of p53 response elements using the crystal structure of p73-DNA complexes and computational studies

Summary: In this study, the molecular details of p73 recognizing response elements (REs) were investigated. The crystal structure and molecular dynamics simulations revealed that p73 recognizes specific nucleotides in different p53 promoter sequences and forms a unique tetramer with each natural promoter sequence through the interaction of key residues. Additionally, the structural features of DNA and the spacing between half-sites influence p73's function.

Summary: This study demonstrates that PIDD1 is recruited to centrosomes by ANKRD26, triggering PIDDosome activation and influencing cell fate. Supernumerary centrosomes resulting from centrosome amplification can activate PIDDosome, and this activation can also occur independently of centrosome amplification in the context of DNA damage.

Inhibitors of ATR and CHK1 enhance damage-induced cell death, while both ATRi and CHK1i can elicit cytotoxicity in some cell lines. Sensitivity is associated with endogenous replication stress, with premature hyper-activation of CDK2 playing a key role in sensitivity to ATRi and CHK1i as single agents.
