An Enhancer-Driven Stem Cell-Like Program Mediated by SOX9 Blocks Intestinal Differentiation in Colorectal Cancer

Xiaoyan Liang et al.

Summary: The study identified SOX9 as a central regulator of an enhancer driven stem cell-like program that hinders intestinal differentiation and promotes CRC development. Ectopic and elevated expression of SOX9 was observed in intestinal adenomas and colorectal adenocarcinomas from mouse models and patients. Disrupting SOX9 activity impaired primary CRC tumor growth by inducing intestinal differentiation.

Yap-Sox9 signaling determines hepatocyte plasticity and lineage-specific hepatocarcinogenesis

Yuchen Liu et al.

Summary: This study elucidates the critical role of Sox9 in YAP-induced cell fate plasticity during hepatocarcinogenesis, particularly in the differentiation of BECs from LPCs. The findings also suggest the importance of YAP activation and SOX9 expression in tumor plasticity in both mouse and human liver tumors.

Integrated analysis of single-cell and bulk RNA sequencing data reveals a pan-cancer stemness signature predicting immunotherapy response

Zhen Zhang et al.

Summary: By analyzing single-cell RNA sequencing data from ICI-treated patients, it was found that cancer stemness may be associated with resistance to ICI treatment. The developed novel stemness signature (Stem.Sig) showed improved predictive performance for ICI response across multiple cancers, potentially serving as a competitive tool for patient selection in immunotherapy.

Unraveling the effect of intra- and intercellular processes on acetaminophen-induced liver injury

M. M. Heldring et al.

Summary: In high dosages, acetaminophen (APAP) can cause severe liver damage, but susceptibility to liver failure varies across individuals and is influenced by factors such as health status. This study aimed to quantify the importance of specific modules in determining the outcome after an APAP insult and identifying potential targets for therapies. The results showed that metabolism and detoxification efficiency, as well as senescence, play crucial roles in necrotic damage and recovery.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

LGR5/R-Spo1/Wnt3a axis promotes stemness and aggressive phenotype in hepatoblast-like hepatocellular carcinoma cell lines

Soheil Akbari et al.

Summary: LGR5 is a newly defined stem cell marker expressed in endoderm-derived organs such as the small intestine, colon, and pancreas. It plays a significant role in liver regeneration and stem cell maintenance, with high expression observed in various cancers including hepatocellular carcinoma. Activation of the canonical Wnt pathway increases LGR5 expression in a subset of HCC cell lines with a "hepatoblast-like" appearance, resulting in enhanced tumor-promoting abilities.

ZNRF3 and RNF43 cooperate to safeguard metabolic liver zonation and hepatocyte proliferation

Summary: Recent research has shown that restricted chromatin accessibility in intestinal stem cells (ISCs) prevents the expression of metabolic enzymes, while fine-tuning of WNT/β-Catenin activity by ZNRF3 and RNF43 restricts proliferation in a subset of hepatocytes, preserving metabolic function. ZNRF3 and RNF43 cooperate to spatially and temporally constrain WNT/β-Catenin activity in the liver, maintaining a balance between metabolic function and hepatocyte proliferation.
