High-speed high-resolution data collection on a 200 keV cryo-TEM

Jared Peck et al.

Summary: This study tested key parameters to increase the data collection speed in single-particle cryo-EM and successfully obtained high-resolution EM maps of mouse apoferritin. By using specific methods and operating procedures, rapid determination of single-particle cryo-EM structures can be achieved.

Structures in G proteins important for subtype selective receptor binding and subsequent activation

Volker Jelinek et al.

Summary: By comparing the influence of subtype-specific G alpha structures on the stability of GPCR-G protein complexes and the activation by two Gq-coupled receptors, key selectivity determinants in G alpha, including the N-terminus, alpha N/beta 1-hinge, beta 2/beta 3-loop, and alpha 5 helix, were identified. These determinants differ in their impact on selective binding to GPCRs and subsequent activation depending on the specific receptor, providing new insights into the molecular basis of G protein-coupling selectivity even beyond the G alpha C-terminus. Jelinek, Mosslein & Bunemann conclude that various regions, especially outside of the C-terminal tail of the G alpha subunit, collectively affect G-protein-coupling selectivity.
