ACK1 Contributes to the Pathogenesis of Inflammation and Autoimmunity by Promoting the Activation of TLR Signaling Pathways

Lina Jing et al.

Summary: This study found that ACK1 is involved in the pathogenesis of inflammation and autoimmune diseases mediated by TLR pathways, presenting a potential target for controlling inflammation and alleviating autoimmune diseases.

Early BCR-ABL1 kinetics are predictive of subsequent achievement of treatment-free remission in chronic myeloid leukemia

Naranie Shanmuganathan et al.

Summary: The study evaluated factors influencing treatment-free remission (TFR) in chronic myeloid leukemia patients, finding that the halving time of BCR-ABL1, BCR-ABL1 transcript type, and duration of TKI exposure before attempting TFR were independent predictors of sustained TFR.

Kinase inhibition in autoimmunity and inflammation

Ali A. Zarrin et al.

Summary: There are still unmet medical needs in the treatment of autoimmune and inflammatory diseases, despite recent advances in the field. Inhibitors targeting the activity of kinases that regulate production of inflammatory mediators have shown promising progress in drug development.

SAAFEC-SEQ: A Sequence-Based Method for Predicting the Effect of Single Point Mutations on Protein Thermodynamic Stability

Gen Li et al.

Summary: SAAFEC-SEQ is a gradient boosting decision tree machine learning method for predicting the change of folding free energy caused by amino acid substitutions. It does not require the 3D structure of the corresponding protein, making it suitable for genome-scale investigations with sparse structural information.

Toll-like receptor signalling in B cells during systemic lupus erythematosus

Simon Fillatreau et al.

Summary: Loss of B cell tolerance to autoantigens in systemic lupus erythematosus (SLE) is driven by TLR7, whereas TLR9 appears to protect against SLE by limiting the stimulatory activity of TLR7. The unique features of Toll-like receptor signaling in B cells implicate it as a therapeutic target in SLE.

Loss of ACK1 Upregulates EGFR and Mediates Resistance to BRAF Inhibition

Zhenyu Ji et al.

Summary: Targeted suppression of BRAF(V600E) by BRAF inhibitors has revolutionized the treatment of metastatic melanoma, but acquired resistance remains a major challenge. In vemurafenib-resistant melanomas, downregulation of ACK1, a protein kinase involved in EGFR turnover, leads to increased EGFR signaling and drug resistance. Inhibition of ACK1-mediated resistance can be reversed by the EGFR inhibitor gefitinib.

Pharmacological targeting PTK6 inhibits the JAK2/STAT3 sustained stemness and reverses chemoresistance of colorectal cancer

Chaoqun Liu et al.

Summary: The study found that PTK6 is overexpressed in CRC tissues and promotes the stemness and chemoresistance of CRC cells by activating the JAK2/STAT3 signaling pathway. Pharmacological inhibition of PTK6 using XMU-MP-2 significantly reduces stemness property and enhances chemosensitivity in CRC cells.

Putting the BRK on breast cancer: From molecular target to therapeutics

Hui Li Ang et al.

Summary: BRK, a non-receptor tyrosine kinase highly expressed in breast carcinomas, has been found to promote cell proliferation, metastasis, and cancer development. Despite the development of numerous inhibitors against BRK, none have progressed into clinical phase. Understanding the successes and challenges of these inhibitor developments is crucial for future improvements of clinically relevant inhibitors.
