Bivalent molecular mimicry by ADP protects metal redox state and promotes coenzyme B12 repair

Harsha Gouda et al.

Summary: This study identifies an unconventional strategy for controlling metal redox state by using bivalent molecular mimicry with ADP to protect against cob(II)alamin overoxidation. ADP induces a conformational change that seals off solvent access, exerting control over the metal oxidation state. This research reveals the importance of metabolites in preserving and recycling a rare, essential metal cofactor.

The Ah Receptor from Toxicity to Therapeutics: Report from the 5th AHR Meeting at Penn State University, USA, June 2022

Gary H. Perdew et al.

Summary: The aryl hydrocarbon receptor (AHR) senses low-molecular-weight molecule signals from environmental exposures, the microbiome, and host metabolism. It plays important roles in host homeostasis, chronic disease development, and responses to toxic insults. Recent research has shown that AHR is a promising target for cancer, metabolic diseases, skin conditions, and autoimmune disease. This meeting aimed to explore the potential therapeutic applications based on our understanding of this receptor.

Single-cell RNA Sequencing Reveals How the Aryl Hydrocarbon Receptor Shapes Cellular Differentiation Potency in the Mouse Colon

Yongjian Yang et al.

Summary: This study utilized single-cell transcriptomics to analyze colonic intestinal crypts from wild-type and intestinal-specific Ahr knockout mice, revealing the important role of Ahr in regulating colon tumorigenesis. Ahr deletion promoted the expression of Foxm1-regulated genes in specific epithelial cell types and secretory cells, and increased the differentiation potency and rate of cell differentiation in stem cells. Furthermore, Ahr deletion disrupted intercellular signaling. This study provides new evidence of the molecular function of Ahr in vivo and has important implications for cancer prevention.

Summary: TCDD induces reprogramming of intermediate metabolism, including repression of genes in both Cbl-dependent carboxylase and Cbl-independent beta-oxidation-like pathways, as well as inhibition of MUT activity. Additionally, TCDD decreases serum Cbl levels and induces the production of itaconate.

Aryl Hydrocarbon Receptor Signaling Synergizes with TLR/NF-κB-Signaling for Induction of IL-22 Through Canonical and Non-Canonical AhR Pathways

Yasuhiro Ishihara et al.

Summary: IL-22 is a key cytokine involved in gut immunity and host defense, primarily produced by activated T cells. Activation of the aryl hydrocarbon receptor (AhR) in bone marrow-derived macrophages (BMM) can induce the expression and production of IL-22, synergistically with Toll-like receptor (TLR) ligands. The study demonstrates that the synergistic and prolonged induction of IL-22 is dependent on the expression of AhR and Nuclear Factor-kappa B (NF-kappa B) member RelB, integrating signals of canonical and non-canonical AhR pathways.
