Production of 1, 3-medium chain-2-long chain (MLM) triacylglycerols by metabolically engineered Escherichia coli

Summary: In this study, a novel approach for the production of MLM-TAGs using engineered Escherichia coli was proposed. By introducing different gene expressions, MLM-TAGs with high content were successfully synthesized. These findings provide a new alternative for the sustainable production of MLM-TAGs.

Harnessing transcription factor Mga2 and fatty acid elongases to overproduce palmitoleic acid in Saccharomyces cerevisiae

Summary: In this study, a Saccharomyces cerevisiae platform strain was engineered for high-level production of palmitoleic acid (POA). The expression of key enzymes in fatty acid metabolism was upregulated by transcription factor Mga2, resulting in increased POA and total unsaturated fatty acid content. Through combinatorial expression optimization, a recombinant strain with a high POA content of 58.5% (v/v) was successfully established, showing the potential for sustainable production of POA or its derivatives.

Hippo pathway regulation by phosphatidylinositol transfer protein and phosphoinositides

Summary: The Hippo pathway is crucial for development, organ size control, and tissue homeostasis, and its dysregulation is linked to cancer. In this study, a marine natural product called microcolin B (MCB) was identified as a Hippo activator that selectively kills YAP-dependent cancer cells. Structural optimization led to the discovery that phosphatidylinositol transfer proteins alpha and beta (PITP alpha/beta) are the direct molecular targets of MCB. The study reveals a previously unknown role of PITP alpha/beta in regulating the Hippo pathway and suggests them as potential therapeutic targets for cancer.
