Clonal Hematopoiesis Is Associated With Higher Risk of Stroke

Plasma Ceramide Levels Are Elevated in Patients With Early Coronary Atherosclerosis and Endothelial Dysfunction

Nadia Akhiyat et al.

Summary: The study showed that elevated plasma ceramide levels were associated with coronary endothelial dysfunction in early coronary atherosclerosis, suggesting that plasma ceramides may serve as potential biomarkers or therapeutic targets for early coronary atherosclerosis in humans.

Coronary microvascular dysfunction is associated with exertional haemodynamic abnormalities in patients with heart failure with preserved ejection fraction

Ali Ahmad et al.

Summary: This study found that coronary microvascular function is inversely associated with filling pressures, particularly during exercise. Both types of coronary microvascular dysfunction are associated with higher filling pressures at peak exercise.

Clonal hematopoiesis of indeterminate potential (CHIP): Linking somatic mutations, hematopoiesis, chronic inflammation and cardiovascular disease

Christopher S. Marnell et al.

Summary: Clonal hematopoiesis of indeterminate potential (CHIP) refers to clonal expansion of hematopoietic stem cells with leukemogenic mutations in individuals without hematologic malignancy. It is associated with a 0.5-1.0% risk of leukemia per year and a two-fold increase in cardiovascular risk. Mutations in specific genes are common in CHIP patients, and the condition is linked to various inflammatory diseases and prognosis of cardiovascular and other conditions.

Coronary Microvascular Dysfunction

Shigeo Godo et al.

Summary: Accumulating evidence over the past few decades has shown that structural and functional abnormalities of coronary microvasculature are highly prevalent and associated with adverse clinical outcomes in patients with various cardiovascular diseases. CMD has been recognized as an important clinical entity, with potential mechanisms including enhanced coronary vasoconstrictive reactivity at the microvascular level.

Summary: This study found that clonal hematopoiesis mutations are common among HF patients with reduced LVEF and are associated with accelerated HF progression, regardless of whether the etiology is ischemic or nonischemic.

TET2 Protects Against Vascular Smooth Muscle Cell Apoptosis and Intimal Thickening in Transplant Vasculopathy

Allison C. Ostriker et al.

Summary: The study found that TET2 is suppressed in CAV and GA, likely mediated by IFN gamma. TET2 helps to protect VSMCs from apoptosis in the context of transplant vasculopathy or IFN gamma stimulation. Enhancing TET2 enzymatic activity with high-dose ascorbic acid reduces VSMC apoptosis and intimal thickening, suggesting that promoting TET2 activity in CAV may be an effective strategy for limiting CAV progression.

Extracellular NLRP3 inflammasome particles are internalized by human coronary artery smooth muscle cells and induce pro-atherogenic effects

Susanne Gaul et al.

Summary: When extracellular NLRP3 inflammasome particles are internalized into human coronary artery smooth muscle cells, they induce pro-inflammatory and pro-atherogenic effects, representing a novel mechanism of cell-cell communication and perpetuation of inflammation in atherosclerosis. Therefore, extracellular NLRP3 inflammasomes could be used to improve the diagnosis of inflammatory diseases and develop new anti-inflammatory therapeutic strategies.
