3D human liver spheroids for translational pharmacology and toxicology

Magnus Ingelman-Sundberg et al.

Summary: Drug development is a challenging and failure-prone process, with liver toxicity being a major cause of safety failures. The current preclinical systems for compound selection are inadequate, necessitating the development of new strategies to increase clinical success rates. Human liver spheroids are becoming increasingly utilized for various analyses and drug development, showing promise as a new standard in translational pharmacology and toxicology.

Analyses of Transcriptomics Cell Signalling for Pre-Screening Applications in the Integrated Approach for Testing and Assessment of Non-Genotoxic Carcinogens

Yusuke Oku et al.

Summary: With the recent rapid advancement of methodological tools, the mechanistic understanding of biological processes leading to carcinogenesis is expanding. New methodologies such as transcriptomics can provide information on non-genotoxic mechanisms of chemical carcinogens for regulatory applications. The OECD expert group is reviewing assays to be integrated into the Integrated Approach to the Testing and Assessment (IATA) of Non-Genotoxic Carcinogens (NGTxC), focusing on pre-screening gene expression changes associated with phenotypic alterations that underlie carcinogenic processes.

Sorafenib reduces steatosis-induced fibrogenesis in a human3Dco-culture model of non-alcoholic fatty liver disease

Guilherme Ribeiro Romualdo et al.

Summary: NAFLD affects approximately 25% of the global population, with NASH presenting a more progressive form of the disease. Research has shown that sorafenib exhibits antifibrotic effects in a 3D in vitro model, indicating its potential clinical use for the treatment of NAFLD/NASH patients.

Carcinogenesis: Failure of resolution of inflammation

Anna Fishbein et al.

Summary: Inflammation in the tumor microenvironment is a hallmark of cancer and is recognized as a key characteristic of carcinogens. The failure of resolution of inflammation in cancer is being understood. Eicosanoids play a critical role in regulating inflammation and its resolution, while non-genotoxic mechanisms such as inflammation and oxidative stress promote genotoxicity and contribute to carcinogenesis.

Assessing chemical carcinogenicity: hazard identification, classification, and risk assessment. Insight from a Toxicology Forum state-of-the-science workshop

Susan P. Felter et al.

Summary: The workshop discussed challenges related to assessing chemical carcinogenicity, focusing on problem formulation, modes-of-action, and dose-response assessment, and proposed methods to address these issues. Scientific limitations of traditional rodent chronic bioassays were acknowledged, as well as knowledge gaps that need to be addressed for the further development and application of new approach methodologies.

Primary Human Hepatocyte Spheroids as Tools to Study the Hepatotoxic Potential of Non-Pharmaceutical Chemicals

Vania Vilas-Boas et al.

Summary: This study investigated the cholestatic potential of non-pharmaceutical chemicals using primary human hepatocytes cultured in 3D spheroids. The results showed that short exposures to cyclosporine-A consistently decreased the cholestatic index, bosentan exhibited hepatotoxicity, and triclosan markedly deregulated genes involved in different types of hepatotoxicity.
