SIRT3/GLUT4 signaling activation by metformin protect against cisplatin-induced ototoxicity in vitro

Increased mitophagy protects cochlear hair cells from aminoglycoside-induced damage

Summary: This study found that aminoglycosides cause hair cell injury by impairing mitophagy, a process that degrades damaged mitochondria. The impaired mitophagy is characterized by blocked recruitment of PRKN to mitochondria and phagophore recognition of damaged mitochondria. Additionally, neomycin disrupts mitophagy by inhibiting Pink1 expression and inducing ATF3 expression. Treatment with a mitophagy activator can rescue neomycin-treated hair cells by increasing mitophagy. Overall, this study suggests that modulating or intervening in mitophagy may have therapeutic potential for aminoglycoside-induced hearing loss.

Macrophage-mediated immune response aggravates hearing disfunction caused by the disorder of mitochondrial dynamics in cochlear hair cells

Summary: Mitochondrial dynamics play a crucial role in maintaining the physiological function of the mitochondrial network. Our study reveals that deficiency of mitochondrial outer membrane proteins FAM73a or FAM73b leads to outer hair cell damage and progressive hearing loss. Abnormal mitochondrial fusion results in increased oxidative stress and apoptosis of hair cells in the early stage. Additionally, activated macrophages and CD4(+) T cells with elevated expression of inflammatory cytokines contribute to the worsened damage of hair cells in mutant mice.

Single-cell transcriptomic atlas of mouse cochlear aging

Summary: By establishing a dynamic single-cell transcriptomic landscape of cochlear aging, we found that loss of proteostasis and elevated apoptosis are the key features of cochlear aging. We also discovered unexpected transcriptional fluctuations in intermediate cells located in the stria vascularis. Additionally, upregulation of the ER chaperone protein HSP90AA1 can mitigate ER stress-induced damages associated with aging, suggesting a potential therapeutic target for delaying age-related hearing loss.
