Exploration of novel hexahydropyrrolo[1,2-e]imidazol-1-one derivatives as antiviral agents against ZIKV and USUV

Ran Chen et al.

Summary: The main goal of this study is to investigate the anti-ZIKV and anti-USUV activity of a new library of compounds and to preliminarily investigate the mechanism of action of the selected hit compounds in vitro. Two potent anti-ZIKV and anti-USUV agents, namely ZDL-115 and ZDL-116, were discovered, both presenting low cytotoxicity, cell-line independent antiviral activity in the low micromolar range and ability of reducing viral progeny production. Future experiments will be focused on compound modification to discover analogues that are more potent and on the clarification of the mechanism of action and the specific drug target.

Antiviral effects of the fused tricyclic derivatives of indoline and imidazolidinone on ZIKV infection and RdRp activities of ZIKV and DENV

Guang-Feng Zhou et al.

Summary: This study focuses on the anti-ZIKV effects of fused tricyclic derivatives and finds that they have potential as drug leads. Some compounds are broad-spectrum inhibitors of both ZIKV and DENV, while others selectively inhibit one of the viruses. Certain compounds are more active against Asian ZIKV strains and silylation enhances their antiviral activity. The study also highlights the importance of structural features in compound efficacy.

Non-volatile acylphloroglucinol components from Eucalyptus robusta inhibit Zika virus by impairing RdRp activity of NS5

Zhai-Wen Yao et al.

Summary: This study evaluated the ability of non-volatile acylphloroglucinols from Eucalyptus leaves to inhibit ZIKV, and found a new symmetrical acylphloroglucinol dimer with significant inhibitory effects. This compound, eucalyprobusone G, showed better efficacy against ZIKV compared to the reference compound ribavirin without inducing cytotoxicity. Further research indicated that eucalyprobusone G impairs viral binding and RdRp activity of NS5, suggesting it could be developed as an anti-ZIKV agent.
