The practicality of this method has been demonstrated in organic synthesis, including the synthesis of key building blocks for (-)-debromoflustramine B.

Ir-Catalyzed Asymmetric Allylic Alkylation of Dialkyl Malonates Enabling the Construction of Enantioenriched All-Carbon Quaternary Centers

Summary: An enantioselective iridium-catalyzed allylic alkylation of malonates with trisubstituted allylic electrophiles to form all-carbon quaternary stereocenters is reported. This reaction proceeds at ambient temperature and enables the preparation of a wide range of enantioenriched products in up to 93% yield and 97% ee. The quaternary products can be readily converted to several valuable building blocks such as vicinal quaternary products and beta-quaternary acids.

Mechanism-based ligand design for copper-catalysed enantioconvergent C(sp3)-C(sp) cross-coupling of tertiary electrophiles with alkynes

Summary: In contrast to the well-established enantioconvergent radical C(sp(3))-C cross-coupling, the coupling of tertiary electrophiles to forge all-carbon quaternary stereocentres remains unexplored. Here, we demonstrate a general copper-catalysed enantioconvergent C(sp(3))-C(sp) cross-coupling using rational design of chiral ligands. This protocol provides a practical platform for the construction of chiral carbon building blocks of interest in organic synthesis.

Asymmetric Allylation by Chiral Organocatalyst-Promoted Formal Hetero-Ene Reactions of Alkylgold Intermediates

The mechanism involves an alkylgold intermediate, and the X-ray crystal structure of the allylgold species reveals its unique stability and reactivity. Additionally, an asymmetric formal hetero-ene reaction of the gold intermediate is enabled with the assistance of a quinine-derived squaramide catalyst, extending the synthetic applications of gold complexes and the versatility of gold catalysis.

Enantioselective Synthesis of Nitriles Containing a Quaternary Carbon Center by Michael Reactions of Silyl Ketene Imines with 1-Acrylpyrazoles

Using a chiral N,N'-dioxide-Co(II) complex, an efficient enantioselective Michael reaction was achieved for the synthesis of nitrile-, aryl-, and dialkyl-bearing carbon centers, with applications in the divergent synthesis of pharmaceuticals and natural products. The through-space dispersion interactions between unbound silyl ketene imines and the 1-acrylpyrazole-bonded catalyst are crucial for facilitating the reactivity and enantioselectivity of the process.

Asymmetric construction of acyclic quaternary stereocenters via direct enantioselective additions of α-alkynyl ketones to allenamides

The development of an efficient chiral phosphoric acid catalyzed direct asymmetric addition method has allowed for the synthesis of acyclic all-carbon quaternary stereocenters with excellent regioselectivities and high enantioselectivities. The mechanism of this reaction involves a novel covalent allyl phosphate intermediate and asymmetric conjugate addition under chiral anion catalysis. Extensive experimental studies have provided access to a wide array of chiral scaffolds with quaternary stereocenters.
