Activatable fluorescent probes for in situ imaging of enzymes

Xiaofeng Wu et al.

Summary: This article summarizes the methods for in situ imaging of enzymes using activatable fluorescent probes, and proposes that fluorescent probes with both activatable and in situ properties will facilitate high-resolution studies of enzymes in living systems.

Activatable Fluorophores for Imaging Immune Cell Function

Lorena Mendive-Tapia et al.

Summary: Optical imaging has become an essential tool for studying biomolecular processes in live systems with unprecedented spatial resolution. In this Account, recent advances in the synthesis and implementation of activatable fluorescent probes for monitoring immune cell activity are summarized. The Account covers the imaging of the three major cell types in the immune system: neutrophils, macrophages, and lymphocytes.

Neutrophils in cancer: heterogeneous and multifaceted

Catherine C. Hedrick et al.

Summary: Neutrophils have emerged as important regulators of cancer, revealing their diverse and plastic contributions in tumor development and growth, influencing positively and negatively depending on time, place, and disease context. Their functional importance in cancer has been overlooked due to their short-lived, terminally differentiated, and non-proliferative characteristics, but recent studies have led to a paradigm shift in understanding their roles.

A Functional Chemiluminescent Probe for in Vivo Imaging of Natural Killer Cell Activity Against Tumours

Jamie I. Scott et al.

Summary: A new chemiluminescent probe has been developed to image the killing activity of NK cells against cancer cells in vivo, demonstrating high selectivity and signal-to-noise ratios. This is the first chemiluminescent probe for in vivo imaging of NK cell activity in live tumors.

The regulatory mechanism of neutrophil extracellular traps in cancer biological behavior

Hui Wang et al.

Summary: Neutrophil extracellular traps (NETs) are a crucial host defense mechanism, playing significant roles not only in infectious inflammation but also in noninfectious conditions. Regulation of NETosis process by various stimuli and signaling pathways can impact the release of NETs, which in turn affects cancer biological behaviors.

A fluorogenic peptide-based smartprobe for the detection of neutrophil extracellular traps and inflammation

Maria R. Rios et al.

Summary: A highly sensitive optical probe for the detection of activated neutrophils and NETs was reported in this study. This probe, based on a triple-quenched, super-silent tri-branched structure, showed a >20 fold increase in fluorescence upon cleavage. It was highly specific for human neutrophil elastase and successfully detected NETosis and neutrophil activation in in vitro differentiated neutrophils and isolated human neutrophils.
