Enantio- and Diastereoselective De Novo Synthesis of 3-Substituted Proline Derivatives via Cooperative Photoredox/Bronsted Acid Catalysis and Epimerization

Chao Che et al.

Summary: A novel strategy for catalytic asymmetric synthesis of enantioenriched 3-cis- and 3-trans-substituted prolines has been successfully developed via a unique cascade radical addition/cyclization enabled by synergistic photoredox/Bronsted acid catalysis and subsequent base-assisted epimerization. This method provides a de novo access to all four stereoisomers of 3-substituted prolines, which cannot be readily obtained using current established methods. The methodology can also be extended to the asymmetric synthesis of the full complement of stereoisomers of 3-substituted pipecolinic acids.

Enantioselective Radical Reactions Using Chiral Catalysts

Shovan Mondal et al.

Summary: Benefiting from the increase in fundamental knowledge, there has been a continuous burst of new ideas and catalytic methods for enantioselective radical reactions in the past 20 years. This review provides a comprehensive survey of the progress achieved during this period, focusing on chiral organocatalysts and transition-metal redox catalysis. The review also discusses the merger of photocatalysis with organo- or metalla-photocatalysis and enzyme-catalyzed reactions involving radical intermediates.

Ketone-Olefin Coupling of Aliphatic and Aromatic Carbonyls Catalyzed by Excited-State Acridine Radicals

Nicholas J. Venditto et al.

Summary: This article introduces a mild and metal-free ketone-olefin coupling reaction using an excited-state acridine radical super reductant as a photoredox catalyst. Both intramolecular and intermolecular ketone-olefin couplings of aliphatic and aromatic ketones and aldehydes are demonstrated, along with mechanistic evidence supporting an olefin-first ketone-olefin coupling mechanism.

Bioinspired o-Naphthoquinone-Catalyzed Aerobic Oxidation of Alcohols to Aldehydes and Ketones

Jisun Baek et al.

Summary: A biomimetic ADH-like oxidation protocol using an ortho-naphthoquinone catalyst and a catalytic amount of base has been developed. The organocatalytic aerobic oxidation proceeds through intramolecular 1,5 hydrogen atom transfer, which is mechanistically distinctive from previous ADH mimics that require metals as catalysts. This ADH-like oxidation protocol provides a green alternative to existing stoichiometric and metal-catalyzed alcohol oxidation reactions.

Asymmetric Oxidative Lactonization of Enynyl Boronates

Kezhuo Zhang et al.

Summary: This study presents a novel strategy for the preparation of gamma-lactones through the oxidation of enynyl boronates. By oxidizing the C(sp)-B bond and subsequently performing epoxidation and lactonization, various gamma-lactones and unsaturated butenolides can be efficiently synthesized. Furthermore, asymmetric oxidative lactonization of enynyl boronates can generate chiral gamma-lactones with high enantioselectivities and diastereoselectivities. The versatile transformations and prevalence of gamma-lactones highlight the importance of this strategy in the construction and late-stage functionalization of complex molecules.

Construction of axially chiral compounds via catalytic asymmetric radical reaction

Dong Liang et al.

Summary: The study of the chemistry of axially chiral compounds has become increasingly important due to their widespread presence in various fields. Catalytic asymmetric radical reactions provide a complementary platform for the construction of enantiomerically enriched compounds, which are in high demand in the chemical and pharmaceutical industries. This review critically discusses recent achievements in this area based on different radical precursors and catalytic activation modes.

Stereodivergent Synthesis of Enantioenriched γ-Butyrolactones Bearing Two Vicinal Stereocenters Enabled by Synergistic Copper and Iridium Catalysis

Lu Xiao et al.

Summary: Utilizing a new reaction model, gamma-butyrolactones with diverse stereoisomers were prepared stereospecifically through a synergistic Cu/Ir-catalyzed asymmetric cascade reaction, allowing control of vicinal stereocenters. The study of the chiral intermediate shed light on the regio- and stereoselectivity of the initial bond formation process, while control experiments provided insight into catalyst interactions in the dual catalytic system. Enantioenriched products were further converted into structurally complex chiral molecules and organocatalysts, expanding the synthetic accessibility of these compounds.

Asymmetric radical carboesterification of dienes

Xiaotao Zhu et al.

Summary: The authors present a method to form chiral esters from conjugated dienes with copper and chiral PyBox ligands, likely proceeding via an allylic radical. This work represents an advance in the enantioselective construction of C-O bond on open-chain hydrocarbon radicals, which may lead to the discovery of other asymmetric radical reactions. The participation of external acids in the reaction substantially extends its applicability and leads to structurally diverse allylic ester products.
