Atroposelective Three-Component Coupling of Cyclic Diaryliodoniums and Sodium Cyanate Enabled by the Dual-Role of Phenol

Summary: A copper-catalyzed atroposelective ring-opening reaction of cyclic diaryliodoniums, sodium cyanate (NaOCN) and phenols is reported. The reaction selectively produces axially chiral carbamates by coupling cyclic diaryliodoniums and NaOCN, followed by reaction with phenols. Mechanistic investigations reveal that phenols serves not only as a reagent to trap highly active intermediate isocyanates, but also as a standby ligand to activate the copper catalyst. The carbamates can be readily transformed into highly functionalized urea derivatives through a simple nucleophilic substitution reaction.

Atroposelective Access to 1,3-Oxazepine-Containing Bridged Biaryls via Carbene-Catalyzed Desymmetrization of Imines

Summary: In this study, a novel bridged biaryl containing medium-sized rings was selectively synthesized via N-heterocyclic carbene organocatalysis. The reaction began with the addition of the carbene catalyst to the aminophenol-derived aldimine substrate, followed by oxidation and intramolecular desymmetrization to form 1,3-oxazepine-containing bridged biaryls with high yields and excellent enantioselectivities. These products can be readily transformed into chiral phosphite ligands.

Dirhodium(II)/Phosphine Catalyst with Chiral Environment at Bridging Site and Its Application in Enantioselective Atropisomer Synthesis

Summary: A dirhodium(II)/phosphine catalyst was developed for the asymmetric arylation of phenanthrene-9,10-diones with arylboronic acids. The chiral environment at the bridging site and tuning of axial and bridging ligands enabled highly enantioselective carbonyl addition reaction. Kinetic analysis showed that the rate-determining step involved the dirhodium(II) complex and arylboronic acid. Axially chiral biaryls were prepared through an oxidative ring-opening reaction.

Summary: Arising from restricted rotation in single bonds due to steric or electronic effects, atropisomerism is a fundamental category for molecules to exhibit their three-dimensional characters, particularly in axially chiral biaryl compounds. Despite the widespread occurrence of axially chiral skeletons in natural products and bioactive molecules, catalytic asymmetric methods for their synthesis face challenges in controlling stereoselectivity for highly sterically hindered variants. However, recent studies have shown successful direct asymmetric synthesis of biaryl atropisomers through transition metal catalysis, offering insights into the activation of substrates and changes in chemoselectivity through torsional strain-promoted ring-opening reactions.

Organocatalytic dynamic kinetic resolution of N-arylindole lactams: atroposelective construction of axially chiral amino acids bearing a C-N chiral axis

Enantioselective alkylative cross-coupling of unactivated aromatic C-O electrophiles

Torsional strain inversed chemoselectivity in a Pd-catalyzed atroposelective carbonylation reaction of dibenzothiophenium

Summary: A palladium-catalyzed enantioselective ring-opening/carbonylation reaction of cyclic diarylsulfonium salts is reported. The sulfonium salts exhibited higher reactivity compared to thioethers, allowing the reaction to be conducted under mild conditions (room temperature). The steric repulsion of non-hydrogen substituents adjacent to the axis induced distortion in cyclic diarylsulfonium salts, leading to a preference for breaking the exocyclic C-S bond in the ring-opening reaction.

This review summarizes key achievements in the stereoselective preparation of biaryl, heterobiaryl, and nonbiaryl atropisomers between 2015 and 2020, emphasizing synthetic strategies for each structural class and potential applications of atropochiral targets.

Catalytic Atroposelective Dynamic Kinetic Resolution of Biaryl Lactones with Activated Isocyanides

Activated isocyanides were used as the reagent to access novel axially chiral oxazole-substituted biaryl phenols in high yields with high enantioselectivities. The key to success lies in the tandem atroposelective addition of isocyanides to the lactone substrate followed by a rapid cyclization to overcome stereochemical leakage induced by lactol formation.

Summary: This tutorial review provides a comprehensive overview of methodologies available for the atroposelective transformation of (heterobiaryl)biaryl precursors towards enantiomerically enriched products and discusses the conceptual aspects associated with each type of transformation. Several strategies have been developed based on the presence or absence of symmetry in the starting material and the occurrence of racemization or dynamicization events, including desymmetrization, classical kinetic resolution (KR), dynamic kinetic resolution (DKR), and dynamic kinetic asymmetric transformation (DYKAT). Seminal contributions and selected examples are discussed to highlight the potential of these synthetic tools.
