OrbiSIMS metrology Part I: Optimisation of the target potential and collision cell pressure

Lidija Matjacic et al.

Summary: The OrbiSIMS instrument combines speed and high-performance mass spectrometry for 3D imaging at the microscale and nanolayers, with applications ranging from organic electronics to drug discovery. We conducted a systematic study on two key parameters, target potential and collision cell pressure, to evaluate the transmission of secondary ion intensities and provide optimization guidance.

Elucidating the molecular landscape of the stratum corneum

Nichola J. Starr et al.

Summary: In this study, the 3D OrbiSIMS technique was used to analyze the molecular chemistry of human skin tissue and provide unprecedented details about the nonhomogeneity of the stratum corneum. The analysis resolved debates about chemical gradients within the epidermis and revealed similarities in epidermal compounds between human and porcine skin. Additionally, the study demonstrated the permeation of an antiaging peptide into the human stratum corneum.

Drug Resistance in Glioblastoma: The Two Faces of Oxidative Stress

Christophe Olivier et al.

Summary: GBM is a common primary brain tumor with poor prognosis, driven by GSCs in specialized microenvironments and characterized by resistance to therapy. ROS play a dual role in controlling cellular stability, affecting cell cycle, progression, and drug resistance in GBM, while also inducing cell death programs like apoptosis and autophagy. The metabolic adaptation and stromal components in the microenvironment contribute to oxidative stress and drug resistance in GBM progression.

Serotonin Pathway in Cancer

Pragathi Balakrishna et al.

Summary: Serotonin plays a significant role as a neurotransmitter in the central nervous system and a motility mediator in the gastrointestinal tract, with ongoing research into its role in tumorigenesis in various cancers. Drugs like octreotide and telotristat are being investigated for their potential in treating neuroendocrine cancers, with serotonin receptor antagonists showing anticancer effects in in vitro studies across different cancer types. Further in vivo studies are needed to fully understand serotonin's role in cancer and its potential as an anticancer therapeutic target.

Sequential Orbitrap Secondary Ion Mass Spectrometry and Liquid Extraction Surface Analysis-Tandem Mass Spectrometry-Based Metabolomics for Prediction of Brain Tumor Relapse from Sample-Limited Primary Tissue Archives

Joris Meurs et al.

Summary: A novel surface mass spectrometry strategy was used to perform untargeted metabolite profiling of pediatric ependymoma samples, detecting 887 metabolites and identifying key metabolites and pathways predictive of tumor relapse through data fusion and multivariate analysis. This sequential mass spectrometry strategy has proven to be a versatile tool for high-throughput metabolite profiling on sample-limited tissue archives.
