Quantitative Single-Molecule Electrochemiluminescence Bioassay

Summary: A single-molecule electrochemiluminescence bioassay is developed for imaging and direct quantification of single biomolecules. This assay localizes the electrochemiluminescence events and allows mapping of biomolecule distribution. By merging repeated reactions and counting clustered molecules, biomolecules can be quantified. The developed bioassay shows a detection limit of 67 attomole concentration and represents a new tool for specific bioimaging and ultrasensitive quantitative analysis.

Plasmon-Boosted Fe, Co Dual Single-Atom Catalysts for Ultrasensitive Luminol-Dissolved O2 Electrochemiluminescence Detection of Prostate-Specific Antigen

Summary: In this study, an efficient luminol-DO ECL system was developed through the complexing of Fe, Co dual single-atom catalysts (D-SACS) supported by N-doped graphene with the luminol-capped Ag nanoparticles (AgNPs). The performance of the corresponding ECL system could be significantly boosted up to 677-fold under optimal testing conditions, comparable to the classic luminol-O-2 system. The developed luminol-DO ECL system was successfully applied for the stable ultrasensitive detection of prostate-specific antigen (PSA) in a wide linear range of 1 fg/mL to mu g/mL, with a low limit of detection (0.98 fg/mL).

The combination of highly efficient resonance energy transfer in one nanocomposite and ferrocene-quenching for ultrasensitive electrochemiluminescence bioanalysis

Summary: In this study, a facile strategy for the design of highly efficient electrochemiluminescence (ECL) sensor was proposed for the sensitive and accurate detection of prostate-specific antigen (PSA). The integration of carboxylated graphitic carbon nitride (g-C3N4) nanosheet (CCN) and tris (2, 2'-Bipyridyl) ruthenium (II) (Ru(bpy)32+) encapsulated in silica nanospheres (RuSi NPs) allowed for a shortened electron-transfer path and increased resonance energy transfer (RET) efficiency. The ECL signal was remarkably decreased through the introduction of aptamer, PSA, and ferroceneboronic acid (FcBA), enabling the ultrasensitive detection of PSA with high selectivity.

Summary: A new biosensing platform for HPV-16 virus detection was developed using PCN-224/nano-zinc oxide composites, cyclic amplification, and hybridization chain reaction. The stability of the electrode was improved by introducing a hybrid substrate material based on PCN-224, nano-zinc oxide, and polyacrylamide. The amplification efficiency of the target HPV-16 was greatly enhanced, and the signal-off detection of HPV was achieved through cycling cleavage and HCR-amplified signal.

Cyclic voltammetry and differential pulse voltammetry of C545T and C545T MRs showed multiple oxidation peaks, indicating that the multiple charge states of C545T could be produced by continuous electron- or hole-injection processes. The ECL system with C545T MRs/TEOA showed intense yellow emission with high selectivity and sensitivity for dopamine detection.

Summary: This study developed a new electrochemiluminescence assay method using two-dimensional black phosphorus nanoflakes for highly sensitive and selective detection of trace lead ions. The method demonstrated effective energy transfer between the BP nanoflakes and Ag/AgCl nanocubes, resulting in enhanced ECL emission in the presence of lead ions. This approach could accurately quantify lead ions in a range from 0.5 pM to 5 nM and presents a promising route for potential applications in clinical analysis.
