Online Collision-Induced Unfolding of Therapeutic Monoclonal Antibody Glyco-Variants through Direct Hyphenation of Cation Exchange Chromatography with Native Ion Mobility-Mass Spectrometry

Guusje van Schaick et al.

Summary: Post-translational modifications (PTMs) increase protein structural heterogeneity and can alter conformation and biological functions. Monitoring PTMs is crucial for therapeutic products like monoclonal antibodies (mAbs). Cation-exchange chromatography (CEX) with native mass spectrometry is valuable for characterizing mAb charge variants, but cannot explore protein conformation. To address this, we combined CEX separation with collision-induced unfolding (CIU) experiments to monitor conformational differences without compromising CEX resolution. This strategy enabled in-depth characterization of mAb glycoforms and linking conformational changes to resistance against gas-phase unfolding charge variants.

Assessing developability early in the discovery process for novel biologics

Monica L. Fernandez-Quintero et al.

Summary: Beyond potency, a good developability profile is essential for a biological drug. Early selection and screening for these attributes can save resources and avoid costly failures. This review highlights key developability properties, such as the source, biophysical and pharmacokinetic properties, and recombinant expression, and discusses methods for identifying molecules with liabilities and optimizing drug leads. The article also compares developability parameters for injectable and orally delivered biologics and provides an outlook on emerging trends in biologics development.

Non-targeted characterization of attributes affecting antibody-FcγRIIIa V158 (CD16a) binding via online affinity chromatography-mass spectrometry

Daniel W. Woodall et al.

Summary: Antibodies facilitate targeted cell killing by engaging with immune cells through Fc gamma receptors. In this study, the binding affinity of the receptor Fc gamma RIIIa V158 for therapeutic antibodies was evaluated, and the importance of glycosylation patterns on the receptors for mediating binding interactions was highlighted.

mD-UPLC-MS/MS: Next Generation of mAb Characterization by Multidimensional Ultraperformance Liquid Chromatography-Mass Spectrometry and Parallel On-Column LysC and Trypsin Digestion

Saban Oezipek et al.

Summary: In this study, a multidimensional ultra-performance-liquid-chromatography-mass spectrometry system was developed for the characterization and quantification of post-translational modifications (PTMs) of therapeutic antibodies. This system allows rapid analysis and decreases the risk of artificial modifications during sample preparation. The introduced system incorporates a novel in-parallel trypsin and LysC on-column digestion setup, enhancing characterization by generating two distinct peptides.

Risk-Based Control Strategies of Recombinant Monoclonal Antibody Charge Variants

Alain Beck et al.

Summary: Since the first approval of muromonab-CD3, an anti-CD3 recombinant monoclonal antibody, mAb therapeutics have become increasingly important. Charge heterogeneity, a typical feature of mAbs, is highly sensitive to process changes and requires risk-based control strategies to maintain consistent product quality.

HDX-MS for Epitope Characterization of a Therapeutic ANTIBODY Candidate on the Calcium-Binding Protein Annexin-A1

Marius Gramlich et al.

Summary: This study identified the interaction site between an ANXA1-specific antibody and ANXA1 using HDX-MS, revealing the binding region within domain III of ANXA1. The antibody-binding region corresponds to the hydrophobic binding pocket of the N-terminal domain in the absence of calcium. This demonstrates the potential of HDX-MS in studying binding regions and defining action profiles of antibodies for therapy.

Multi-dimensional LC-MS: the next generation characterization of antibody-based therapeutics by unified online bottom-up, middle-up and intact approaches

Julien Camperi et al.

Summary: The accelerated development of new therapeutics in a competitive landscape necessitates high throughput analytical platforms. Emerging formats of biotherapeutics have increased the complexity, emphasizing the need to enhance conventional liquid chromatography resolution. Liquid chromatography-mass spectrometry technologies, such as multi-attribute methods, show potential in complementing traditional analytical toolbox for monoclonal antibody characterization. Multi-dimensional LC-MS provides a promising approach to address the challenges faced with novel antibody formats by combining sample preparation and multi-level analysis in a single chromatographic setup.
