Advances in nucleic acid amplification techniques (NAATs): COVID-19 point-of-care diagnostics as an example

Tongjia Kang et al.

Summary: Achieving superhigh sensitivity is the ultimate goal in modern bio-detection. Nucleic acid amplification technologies (NAATs) are revolutionizing the field, offering novel methods for diagnosis with high efficiency and cost-effectiveness. This review discusses the principles, recent advancements, strengths, and challenges of different NAATs, with a focus on rapid and accurate point-of-care diagnosis. The application of these technologies in the context of the COVID-19 pandemic is also explored, emphasizing the significance of NAATs in combating the disease. Furthermore, the potential for enhancing sensitivity and specificity in biodetection through NAATs is discussed, aiming to fully exploit the capabilities of each technique.

Clinical Validation of a Rapid Variant-Proof RT-RPA Assay for the Detection of SARS-CoV-2

Dounia Cherkaoui et al.

Summary: The COVID-19 pandemic has highlighted the need for expanding diagnostic capabilities to accommodate high-volume testing during peak demand. A rapid nucleic acid testing method based on isothermal amplification has been validated as an alternative to real-time reverse transcription polymerase chain reaction (RT-PCR), helping to address the current testing gap. This clinical validation study demonstrates the effectiveness of a one-pot multi-gene reverse transcription recombinase polymerase amplification (RT-RPA) assay in detecting SARS-CoV-2 variants, while also achieving high sensitivity and specificity with clinical samples. The assay showed no cross-reactivity with other respiratory pathogens and successfully detected all tested SARS-CoV-2 lineages, including variants of concern.

Microbiological Laboratory Diagnosis of Human Brucellosis: An Overview

Giovanni Di Bonaventura et al.

Summary: Brucella spp. are responsible for brucellosis, a common zoonotic infection in specific geographic areas with underreported cases. Laboratory diagnosis is crucial for human cases, with culture, serological tests, and PCR-based methods being the main approaches. Despite advancements in serological tests and NAATs, culture remains the gold standard due to clinical relevance. Further development of NAATs is needed before commercialization for brucellosis diagnosis.
