Disease modeling following organoid-based expansion of airway epithelial cells

Evelien Eenjes et al.

Summary: The study investigated the use of organoid expansion to establish air-liquid interface (ALI) cultures from clinical samples with low epithelial cell numbers. Cells from different sources and expanded in different ways showed variations in cellular composition, response to cigarette smoke exposure, and differentiation modulation. Organoid-based expansion proved to be a valid method for establishing ALI cultures from samples with low cell numbers.

Summary: Studies have shown that premature cells exhibit abnormal differentiation and mitochondria quality control, lower sensitivity to oxygen consumption, and declining ciliary function, indicating increased sensitivity to environmental stressors.

CPAP protects against hyperoxia-induced increase in airway reactivity in neonatal mice

Peter M. MacFarlane et al.

Summary: The study found that CPAP and mild hyperoxia exposure separately increased airway reactivity in neonatal mice, but the combination of CPAP and hyperoxia no longer induced airway hyperreactivity. The combined treatment normalized smooth muscle and epithelial proliferation to control values. The interaction between CPAP-induced stretch and mild hyperoxia exposure has important implications for airway pathophysiology in former preterm infants.
