Functional Evaluation of Human Bioengineered Cardiac Tissue Using iPS Cells Derived from a Patient with Lamin Variant Dilated Cardiomyopathy

Koichiro Miura et al.

Summary: This study investigates the function of cardiac cell sheet tissue derived from patients with lamin variant dilated cardiomyopathy. The results show that the cardiac cell sheet tissue with lamin variant exhibits decreased contractile force and maximum contraction velocity. mRNA expression analysis also reveals downregulation of certain contractile proteins, cardiac transcription factors, Ca2+-handling genes, and ion channels. The bioengineered cardiac tissue with the lamin variant mutation may serve as a promising tissue model for understanding the underlying mechanisms of dilated cardiomyopathy.

Nuclear lamin isoforms differentially contribute to LINC complex-dependent nucleocytoskeletal coupling and whole-cell mechanics

Amir Vahabikashi et al.

Summary: The interactions between the nucleus and cytoskeleton play a crucial role in regulating cellular mechanics. A-type lamins engage with F-actin and vimentin intermediate filaments to modulate cortical stiffness and contractility, while B-type lamins predominantly interact with vimentin intermediate filaments to regulate cytoplasmic stiffness and contractility. The loss of each lamin isoform affects nucleocytoskeletal interactions and cellular mechanics.

Actin-microtubule cytoskeletal interplay mediated by MRTF-A/SRF signaling promotes dilated cardiomyopathy caused by LMNA mutations

Caroline Le Dour et al.

Summary: Mutations in the LMNA gene cause dilated cardiomyopathy and increased ERK1/2 activity in the heart. This study demonstrates that the interaction between cofilin-1 and MRTF-A/SRF mediates the interplay between actin and microtubule cytoskeletons, promoting dilated cardiomyopathy caused by LMNA mutations.

Summary: Laminopathies are rare disorders caused by mutations in the LMNA gene, which can affect multiple systems. While the exact pathomechanisms are not fully understood, various therapeutic approaches have been evaluated, particularly focusing on treatments for striated muscle laminopathies and premature ageing syndromes.

Summary: Mutations in the LaminA gene are a common cause of monogenic dilated cardiomyopathy. In mice, cardiomyocyte-specific expression of the SUN1 miniprotein can extend lifespan to more than one year, showing potential as a therapeutic option for treating this disease.
