Single-cell multiomics reveals increased plasticity, resistant populations, and stem-cell-like blasts in KMT2A-rearranged leukemia

Changya Chen et al.

Summary: The study revealed distinct characteristics of leukemia cells in KMT2A-r ALL patients of different ages. In younger patients, leukemia cells showed increased lineage plasticity, downregulation of steroid response pathways, and the presence of an HSPC-like population. These findings provide new insights into how leukemia cells in young patients evade treatment and immune control.

Lineage Conversion in Pediatric B-Cell Precursor Acute Leukemia under Blinatumomab Therapy

Alexandra Semchenkova et al.

Summary: We reported the incidence and molecular characteristics of lineage switch in 182 pediatric patients with B-cell precursor acute lymphoblastic leukemia (BCP-ALL) treated with blinatumomab. We identified six cases of lineage switch occurring after or during blinatumomab exposure. This study highlights the importance of investigating clonal architecture and molecular changes in identifying lineage switch in BCP-ALL patients.

CD5-positive marginal zone lymphoma: Clinicopathological features and survival outcomes

Summary: CD5 expression in marginal zone lymphoma is not relevant to the prognosis of the patients, but CD5-positive marginal zone lymphoma seems more likely to transform into diffuse large B-cell lymphoma.

Single-cell transcriptomics reveals a distinct developmental state of KMT2A-rearranged infant B-cell acute lymphoblastic leukemia

Eleonora Khabirova et al.

Summary: This study investigates the developmental state of KMT2A-rearranged infant B-ALL and reveals its unique features compared to other childhood B-ALLs. The research shows that this leukemia harbors hybrid myeloid-lymphoid characteristics and suggests potential targetable antigens for specific cancer treatment.

DUX4r, ZNF384r and PAX5-P80R mutated B-cell precursor acute lymphoblastic leukemia frequently undergo monocytic switch

Michaela Novakova et al.

Summary: The study documented a BCP-ALL subtype with early switch to monocytic lineage and loss of B-cell immunophenotype, correlated with three genetic subtypes. The monocytic switch had significant impact on residual disease assessment and treatment outcomes.

Single-cell profiling identifies pre-existing CD19-negative subclones in a B-ALL patient with CD19-negative relapse after CAR-T therapy

Tracy Rabilloud et al.

Summary: The study demonstrates that CD19-negative leukemic subclones may already exist before CAR-T cell therapy in patients with B-ALL, leading to potential relapse. By utilizing single-cell RNA sequencing, pre-existing CD19 negative subclones were identified, highlighting the possibility of assessing the risk of targeted therapy failure.

Cell Lineage Infidelity in PDAC Progression and Therapy Resistance

Antonia Malinova et al.

Summary: Infidelity to cell fate can lead to differentiated cells losing their original identity, which can potentially benefit wound repair but can also be exploited by cancer cells for disease progression. In pancreatic ductal adenocarcinoma, different molecular subtypes reflect varying degrees of fidelity to pancreatic endodermal precursors.

Lineage Switch in an Infant B-Lymphoblastic Leukemia With t(1;11)(p32;q23); KMT2A/EPS15, Following Blinatumomab Therapy

Jing Du et al.

Summary: A 6-month-old infant girl with KMT2A/EPS15-rearranged B-ALL experienced a lineage switch to AML following blinatumomab treatment, achieving remission of both B-ALL and AML by flow cytometry. Despite allogeneic hematopoietic stem cell transplant, she relapsed with CD19+ B-ALL one month post-transplantation. ClonoSEQ(R) analysis revealed the presence of the same clones throughout her disease course, suggesting cellular reprogramming and differentiation.

Wenjuan Liao et al.

Summary: CAR T cell therapy has shown success in achieving complete remission in MLL-r B-ALL patients, but relapse with CD19-negative disease is a common mechanism, with MLL-r patients more prone to developing AML.

CD5+diffuse large B-cell lymphoma: a narrative review

Urshila Durani et al.

Summary: CD5+ DLBCL is a rare subset associated with increased age, high lactate dehydrogenase, B symptoms, extra-nodal sites, higher International Prognostic Index score, and advanced stage in some studies. It is more likely to express Bcl-2, MYC, and MUM1, with a large proportion exhibiting an activated B-cell (ABC)-like phenotype. Studies generally support an independent prognostic value of CD5 in DLBCL, although evidence for more aggressive treatment approaches for CD5+ DLBCL is lacking.

How to Diagnose and Treat CD5-Positive Lymphomas Involving the Spleen

Jose Cabecadas et al.

Summary: Patients with CD5-expressing lymphomas presenting with splenomegaly are often diagnosed with chronic lymphocytic leukemia, but differential diagnosis is important to distinguish from mantle cell lymphoma. Other small B-cell neoplasms involving the spleen and occasionally expressing CD5 include splenic marginal zone lymphoma, hairy cell leukemia, and lymphoplasmacytic lymphoma. The frequency of CD5 positivity varies depending on detection methods, and a combination of morphological, immunophenotypic, and molecular findings is essential for accurate sub-classification of CD5-positive, low-grade B-cell lymphomas of the spleen.
