Research progress of PPARγ regulation of cholesterol and inflammation in Alzheimer's disease

Summary: PPARs are key sensors and regulators of cell metabolism, involved in lipid, glucose and amino acid metabolism, and play important roles in cell differentiation, apoptosis, inflammation regulation, and nerve damage. PPARγ, expressed in different brain regions, regulates lipid metabolism, oxidative stress, and cell apoptosis. It has anti-inflammatory activity and neuroprotection. This article explores the relationship between cholesterol, inflammation, and Aβ in Alzheimer’s disease, and elaborates on the regulation of PPARγ on key proteins and molecules, providing new insights for AD treatment.

Thus, PPAR-gamma agonists play an important role in the inflammatory response of AD or DM patients, and relevant metabolic indices should be considered in clinical therapeutics.

Evidence of epistasis in regions of long-range linkage disequilibrium across five complex diseases in the UK Biobank and eMERGE datasets

Summary: Leveraging linkage disequilibrium (LD) patterns can help discover association signals in genome-wide association studies (GWASs) and investigate non-additive genetic interactions. In this study, we tested the hypothesis that uncovering long-range interactions between loci with strong LD due to epistatic selection can elucidate the genetic mechanisms underlying common diseases. Through association testing, we identified significant and near-significant associations between 23 common diseases and epistatic SNP-SNP pairs in long-range LD. These findings support the role of epistatic interactions in diverse clinical mechanisms and phenotypic outcomes in common diseases.
