NF-κB perturbation reveals unique immunomodulatory functions in Prx1+ fibroblasts that promote development of atopic dermatitis

Kang Ko et al.

Summary: The study reveals the important role of a specific subpopulation of Prx1(+) fibroblasts in immune regulation of the skin. Disruption of the Ikkb-NF-kappa B pathway in these fibroblasts triggers skin inflammation and results in overexpression of CCL11, leading to eosinophil infiltration and T(H)2 immune response, resembling human atopic dermatitis. Monoclonal antibody treatment targeting CCL11 shows promising efficacy in a mouse model.

Spatial transcriptomics of dorsal root ganglia identifies molecular signatures of human nociceptors

Diana Tavares-Ferreira et al.

Summary: Nociceptors, specialized sensory neurons, play a crucial role in detecting and generating pain signals. This study used spatial transcriptomics to characterize the transcriptomes of human nociceptors, identifying different clusters and potential drug targets in the human dorsal root ganglia (DRG). Comparison with mouse sensory neuron transcriptomes and nonhuman primate DRG neuronal subtypes revealed conserved gene expression patterns but also species-specific differences. Sex differences in DRG subpopulation transcriptomes were also identified. This comprehensive spatial characterization of human nociceptors has significant implications for the development of improved treatments for pain disorders.

Environment-dependent alterations of immune mediators in urban and rural south African children with atopic dermatitis

Nonhlanhla Lunjani et al.

Summary: This study found that children with AD had elevated levels of certain immune markers regardless of their location, while children living in rural areas had higher levels of various cytokines compared to urban children. Factors such as early life nutrition, medications, and environmental exposures were associated with differences in circulating cytokine levels in children with AD.

Single-cell analysis reveals innate lymphoid cell lineage infidelity in atopic dermatitis

Natalia Alkon et al.

Summary: This study investigates the phenotypic and molecular features of cutaneous innate lymphoid cells (ILCs) to better understand the biological role of these cells. The findings demonstrate the flexibility of the skin immune system, with ILCs in diseased skin expressing genes characteristic of different immune types. This suggests that the skin immune system can adjust its function to adapt to the environment.

Developmental cell programs are co-opted in inflammatory skin disease

Gary Reynolds et al.

Summary: The skin establishes a complex cellular network early in embryonic development to provide biophysical and immunological protection. The study found an enrichment of innate immune cells in skin during the first trimester, as well as clonal expansion of disease-associated lymphocytes in atopic dermatitis and psoriasis. Additionally, a reemergence of prenatal vascular endothelial cell and macrophage cellular programs was observed in atopic dermatitis and psoriasis lesional skin.

Histamine Increases Th2 Cytokine-Induced CCL18 Expression in Human M2 Macrophages

Susanne Mommert et al.

Summary: The study investigated the role of histamine in CCL18 production in human monocyte-derived M2 macrophages, finding that IL-10 upregulated H2R and increased CCL18 expression through H2R stimulation. This effect was similar in cells from atopic dermatitis patients compared to healthy controls.

Advances in atopic dermatitis in 2019-2020: Endotypes from skin barrier, ethnicity, properties of antigen, cytokine profiles, microbiome, and engagement of immune cells

Takashi Nomura et al.

Summary: Key research advances in atopic dermatitis reveal the complexity of its endotypes, including traditional TH2-dominant type and an axis of cytokines commonly seen in Asian individuals. Innate lymphoid cells play a critical role in AD development and heterogeneity. Stratification based on endotypes may lead to customized therapeutic strategies in the future.

Early-life inflammation primes a T helper 2 cell-fibroblast niche in skin

Ian C. Boothby et al.

Summary: Early-life inflammation can lead to lasting changes in the immune system that confer disease protection or susceptibility. In a mouse skin study, neonatal inflammation was found to cause dysregulation of subcutaneous tissue, with the accumulation of T(H)2 cells and interactions with fibroblasts. This study also identified a T(H)2 cell niche in skin and disease-associated fibroblast populations, suggesting a mechanism of immunological priming through early-life inflammation.

Spontaneously Resolved Atopic Dermatitis Shows Melanocyte and Immune Cell Activation Distinct From Healthy Control Skin

Katharina Rindler et al.

Summary: The study found that the overall cell composition and proteomic profiles of spontaneously healed atopic dermatitis (AD) patients were comparable to healthy controls, without the upregulation of typical AD activity markers. Melanocytes and conventional T-cells in spontaneously healed AD skin may play a role in providing a specific regulatory environment.

Electrical impedance spectroscopy for the characterization of skin barrier in atopic dermatitis

Arturo O. Rinaldi et al.

Summary: This study successfully established a method to directly assess the in vivo status of the epithelial barrier using electrical impedance spectroscopy, demonstrating its usefulness in patients with atopic dermatitis. Electrical impedance spectroscopy showed a significant negative correlation with transepidermal water loss, and a higher sensitivity in discriminating between lesional and non-lesional skin in patients with atopic dermatitis.

Single-cell analysis defines a pancreatic fibroblast lineage that supports anti-tumor immunity

Colin Hutton et al.

Summary: This study used mass cytometry to analyze the stromal composition in murine tissues and tumors, revealing extensive stromal heterogeneity and coordinated relationships between mesenchymal and immune cell subsets in pancreatic ductal adenocarcinoma. The research identified two stable and functionally distinct pancreatic fibroblast lineages marked by CD105 expression, with CD105-positive fibroblasts promoting tumor growth and CD105-negative fibroblasts suppressing tumors, dependent on adaptive immunity. These findings highlight the importance of mesenchymal and immune cell interactions in restricting tumor growth.

Summary: The study introduces a method called sci-Space, which allows for spatial heterogeneity analysis at larger scales while retaining single-cell resolution. By applying this method to developing mouse embryos, the researchers identified thousands of genes exhibiting anatomically patterned expression and leveraged spatial information to annotate cellular subtypes. The results show substantial variation in spatial patterning among cell types and reveal correlations between pseudotime and the migratory patterns of differentiating neurons.

Single-cell RNA-seq reveals fibroblast heterogeneity and increased mesenchymal fibroblasts in human fibrotic skin diseases

Cheng-Cheng Deng et al.

Summary: This study examined fibroblast heterogeneity in keloid, a representative of fibrotic skin diseases, using single-cell RNA-seq. The results revealed that keloid fibroblasts could be divided into 4 subpopulations, with an increased percentage of mesenchymal fibroblasts playing a crucial role in collagen overexpression. This increased mesenchymal fibroblast subpopulation was also found in another fibrotic skin disease, scleroderma, suggesting a broad mechanism for skin fibrosis.
