Efficacy and Safety of Drugs for Gastroparesis: Systematic Review and Network Meta-analysis

Summary: This study evaluated the efficacy and safety of drugs for gastroparesis through a network meta-analysis. The results showed that oral dopamine antagonists and tachykinin-1 antagonists were more effective than placebo. For individual symptoms, oral metoclopramide showed good efficacy for nausea, fullness, and bloating. However, the confidence in the evidence for most comparisons was low to moderate, indicating a need for more effective therapies for gastroparesis.

Neural networks involved in nausea in adult humans: A systematic review

Summary: Nausea is a common clinical symptom that lacks effective treatment with anti-emetic drugs. This study systematically reviewed brain imaging in individuals experiencing nausea in order to identify potential therapeutic targets. The findings suggest that the inferior frontal gyrus, anterior cingulate cortex, and anterior insula are commonly activated during nausea, with some evidence for lateralization. The study also found increased functional connectivity between different cortical regions during nausea. However, limitations include a lack of studies and inconsistent definitions and measurements of nausea.

Clinical trial: a single-centre, randomised, controlled trial of tradipitant on satiation, gastric functions, and serum drug levels in healthy volunteers

Summary: This study aimed to evaluate the effects of Tradipitant on gastric motor functions, satiation, postprandial symptoms, and pharmacokinetics. The experiment conducted on 24 healthy volunteers showed that Tradipitant at a dosage of 85mg b.i.d. for 9 consecutive days did not have a significant impact on gastric emptying, gastric volumes, satiation, or symptoms 30 minutes after a meal. These results are important in relation to the potential use of Tradipitant in patients with gastroparesis.
