Ammonia induces amyloidogenesis in astrocytes by promoting amyloid precursor protein translocation into the endoplasmic reticulum

Summary: Hyperammonemia may be linked to the development of Alzheimer's disease by increasing the amount of mature amyloid precursor protein (mAPP) in astrocytes. This leads to enhanced endocytosis of mAPP and subsequent production of amyloid beta (A beta). Ammonia-induced A beta production in astrocytic endoplasmic reticulum is specifically related to A beta 42, a key component of senile plaques in AD patients.

Summary: This study utilized a multiomics approach to analyze molecular changes during hyperammonemia in muscle tissues, revealing various types of responses including mitochondrial and oxidative dysfunction, disruption of protein homeostasis, and activation of oxidative stress pathways. Additionally, novel observations were made, such as an increase in expression of antiapoptotic proteins, increased calcium flux, and increased protein glycosylation during hyperammonemia. The data demonstrate complex molecular interactions during adaptive and maladaptive responses to cellular stress induced by hyperammonemia.
