Twist1 regulates macrophage plasticity to promote renal fibrosis through galectin-3

Summary: This study reveals a significant association between Twist1 expression in macrophages and the severity of renal fibrosis in IgA nephropathy patients and mice with unilateral ureteral obstruction. Twist1 ablation in macrophages alleviates renal tubular injury and fibrosis, accompanied by reduced macrophage infiltration and M2 polarization in the kidney. Twist1 downregulation inhibits macrophage chemotaxis and migration, at least partially through the CCL2/CCR2 axis. This study highlights the regulation of macrophage plasticity through Twist1/galectin-3 signaling and its potential as a new strategy for delaying kidney fibrosis in chronic kidney disease patients.

The functional multipotency of transforming growth factor β signaling at the intersection of senescence and cancer

Summary: The transforming growth factor beta (TGF-beta) family of cytokines plays a crucial role in regulating various physiological and pathophysiological processes, including immune suppression, tissue remodeling, and cancer progression. TGF-beta 1, in particular, is a well-known and highly active member of this family. It is involved in modulating aging, cellular senescence, and their interaction with cancer. This study reviewed the literature on the role of different TGF-beta family members in the complex interplay between senescence and cancer, highlighting the interaction between normal peritoneal cells and ovarian cancer cells as a central focus. Examples of TGF-beta activity in other normal and cancer cell types were also discussed.

Emerging roles of circRNAs in mice kidney with aging

Summary: In this study, the researchers identified circRNA in the kidneys using RNA sequencing, and constructed a circRNA-miRNA-mRNA interaction network. The results showed that circNpas2 plays an important role in kidney aging, and the ER-related signaling pathway is involved in the regulation. Knockdown of circNpas2 led to cell cycle arrest and inhibition of ER-related protein expression.

Summary: The latest progress in renal fibrosis research reveals the importance of partial epithelial-mesenchymal transition (pEMT) and fatty acid oxidation (FAO) dysfunction. Cpt1a-mediated FAO dysfunction is essential for renal tubular epithelial (RTE) cells undergoing EMT, and rhein inhibits EMT by promoting Cpt1a-mediated FAO. Importantly, FAO depression occurs before EMT and EMT is one of the results of FAO depression.

The results showed that both the fatty acids and oxidized lipoproteins significantly altered the lipid content and composition of LDs. The accumulation of oxidized lipids and dysregulation of lipid metabolism in LDs may be potential targets for the diagnosis and treatment of chronic kidney disease (CKD).
