Artificial intelligence aids in development of nanomedicines for cancer management

Checkpoint Nano-PROTACs for Activatable Cancer Photo-Immunotherapy

Summary: This study reports the use of checkpoint nano-PROTACs combined with photodynamic tumor regression and immunosuppressive protein degradation to block checkpoint signaling pathways for activatable cancer photo-immunotherapy. The nano-PROTACs consist of a photosensitizer and an SHP2-targeting PROTAC peptide connected by a caspase 3-cleavable segment. By degrading SHP2, these nano-PROTACs can block immunosuppressive checkpoint signaling pathways and enhance antitumor immune response. This study represents a generalized PROTAC platform to modulate immune-related signaling pathways for improved anticancer therapy.

Enzyme-triggered deep tumor penetration of a dual-drug nanomedicine enables an enhanced cancer combination therapy

Summary: Emerging nanomedicines can eliminate cancer cells by promoting excessive intracellular reactive oxygen species (ROS) generation. However, tumor heterogeneity and poor penetration of nanomedicines often result in varying levels of ROS production, where low levels of ROS can actually promote tumor cell growth, reducing the effectiveness of these nanomedicines. In this study, a novel amphiphilic and block polymer-dendron conjugate-derived nanomedicine (GFLG-DP/Lap NPs) was developed, incorporating a photosensitizer (Ppa) for ROS therapy and Lapatinib (Lap) for molecular targeted therapy. By combining ROS therapy with Lap, the synergistic effect effectively kills cancer cells, even in deep tumor regions.

Additionally, this conjugate disrupts cell homeostasis, making tumor cells more susceptible to photodynamic therapy.
