Immunogenic cell stress and death

Guido Kroemer et al.

Summary: Immunogenic cell death (ICD) plays an important role in therapy and disease, as dying mammalian cells release signals that interact with the host to determine the immunological correlates of cellular stress and death. ICD is crucial in immunosurveillance and is related to strategies evolved by pathogens and cancer cells. Additionally, normal cell death can also initiate antigen-specific immune responses.

Harnessing cytokines and chemokines for cancer therapy

David J. Propper et al.

Summary: Cytokines play a crucial role in cancer development and progression. While cytokines have been studied for their therapeutic potential in oncology, clinical trials have shown limited efficacy. However, the advent of immunotherapies and a better understanding of the tumor microenvironment have opened up new possibilities for harnessing cytokine networks in cancer treatment.

Nanoscale CaH2 materials for synergistic hydrogen-immune cancer therapy

Fei Gong et al.

Summary: In this study, CaH2 nanoparticles were prepared through liquid-phase exfoliation and used as an antitumor therapeutic agent. When injected into the tumor, the nano-CaH2 reacts with water to produce hydrogen gas, calcium ions, and hydroxyl ions, which enable hydrogen therapy, tumor calcification, and neutralization of the acidic tumor microenvironment. The results showed that the injectable nano-CaH2 dispersion significantly inhibited tumor growth and activated the immune system to promote immune cell infiltration into the tumor, resulting in robust immune responses to inhibit distant tumors. Furthermore, nano-CaH2 dispersed in lipiodol showed improved therapeutic outcomes in interventional transarterial embolization therapy compared to lipiodol alone in a liver cancer model.

Multifunctional metal-organic framework-based nanoreactor for starvation/oxidation improved indoleamine 2,3-dioxygenase-blockade tumor immunotherapy

Liangliang Dai et al.

Summary: This study reports on the co-delivery of glucose oxidase and IDO inhibitor using a metal-organic framework-based nanoreactor, showing an amplified release for tumor starvation/oxidation immunotherapy. The nanosystem overcomes biobarriers associated with tumor penetration, rapidly disassembles and releases cargoes in response to intracellular reactive oxygen species. The combined IDO-blockade immunotherapy enhances immune response and inhibits tumor growth significantly.

Connecting Calcium-Based Nanomaterials and Cancer: From Diagnosis to Therapy

Shuang Bai et al.

Summary: Calcium signaling, as a crucial cellular messenger, plays a role in regulating numerous physiological processes. The concentration of calcium ions is strictly controlled due to their importance, and abnormal regulation of calcium signals can lead to diseases. However, artificial regulation of calcium homeostasis in specific areas may have therapeutic benefits. Calcium overload, characterized by excessive intracellular calcium, can cause cell death but also offers a new approach for cancer treatment. Calcium is involved in various cancer diagnostic and therapeutic events.
