Enhancing the osteogenic differentiation of aligned electrospun poly (L-lactic acid) nanofiber scaffolds by incorporation of bioactive calcium silicate nanowires

Summary: Bone defects cause significant physical and economic burden, but the use of artificially bone repairing materials shows promise. Electrospinning is a popular technique to fabricate fibers for scaffold structures that mimic the extracellular matrix and promote cell attachment and migration. Research has demonstrated the benefits of bioactive ions and physical cues in bone regeneration. In this study, electrospun scaffolds made of calcium silicate and poly (L-lactic acid) were prepared, and the ones with aligned structure exhibited improved physical properties and enhanced cell adhesion and osteogenic gene expression. These findings suggest that the prepared scaffolds have potential for bone regeneration.

Nanoparticles functionalized with stem cell secretome and CXCR4-overexpressing endothelial membrane for targeted osteoporosis therapy

Summary: This study proposes a method using cell-mimicking and targeted therapeutic nanoparticles to replace stem cells. The results show that in a rat model of osteoporosis, this treatment can accumulate in bone, inhibit osteoclast differentiation, and promote osteogenic proliferation. Additionally, this treatment reduces bone mass attenuation in rats with ovariectomy-induced bone loss.

Impairment of 7F2 osteoblast function by simulated partial gravity in a Random Positioning Machine

Summary: Reduced gravity has adverse effects on osteoblast function, and the effects of partial gravity are not well understood. Using a random positioning machine, we simulated partial gravity levels relevant to Mars, Moon, and microgravity conditions. We found that decreasing gravity levels led to reduced cell proliferation and alkaline phosphatase activity, and increased inhibition of mineralization in a dose-dependent manner. The expression of key osteogenic genes was significantly inhibited under Mars-simulating partial gravity, with no further reduction under simulated Moon or microgravity conditions.

Loss of PLC gamma 2 in these cells resulted in changes in macrophage surface markers and phenotype, reduced phagocytic activity and survival, as well as deficits in cellular adhesion and migration.

Aligned and Oriented Collagen Nanocomposite Fibers as Substrates to Activate Fibroblasts

Summary: This study successfully fabricated collagen hydrogels with aligned and oriented fibers doped with gold nanoparticles and carbon nanotubes using a microfluidic device. The doped hydrogels exhibited greater rigidity and stiffness compared to native collagen, with the stiffest formulations found in the presence of carbon nanotubes. In vitro experiments with NIH-3T3 fibroblasts showed directional cell growth on carbon nanotube-doped collagen fibers, as well as increased fibroblast proliferation and secretion of TGF-beta 1.
