Nanopore-Based Protein Identification

Mazdak Afshar Bakshloo et al.

Summary: This study demonstrates a method for protein detection and classification using a nanometer-scale pore formed by the protein aerolysin, achieving detection at the single-molecule level. Different water-soluble proteins treated with the same protease produce different polypeptide fragments, which can be distinguished based on their impact on the nanopore's ionic current, consistent with predicted fragments.

Label-Free Optical Analysis of Biomolecules in Solid-State Nanopores: Toward Single-Molecule Protein Sequencing

Yingqi Zhao et al.

Summary: Sequence identification of peptides and proteins is crucial for proteomics, but current methods face challenges in discriminating amino acids, unfolding proteins, and controlling motion of proteins with nonuniformly charged sequences. Label-free optical analysis techniques, such as localized surface plasmon resonance detection and surface-enhanced Raman spectroscopy on plasmonic nanopores, show promise for addressing these challenges and enabling single-molecule protein sequencing.

Kaikai Chen et al.

Summary: This study introduces a high-resolution nanopore-based DNA sequence mapping method, which successfully labels specific sequence motifs using DNA methyltransferase and oligodeoxynucleotides to achieve high resolution mapping. The article also describes a method for accurately locating the positions of sequence motifs on DNA using nanopore data analysis. This approach enables efficient DNA species identification in an all-electrical format.

Ultrathin Porous Hydrocarbon Membranes Templated by Nanoparticle Assemblies

Grayson L. Jackson et al.

Summary: By using electron beam irradiation of gold or Fe3O4 nanoparticle monolayer sheets followed by wet chemical etching, it is possible to template 2D porous hydrocarbon membranes and transform gold nanoparticle supercrystals into 3D porous hydrocarbon membranes. The control of nanoparticle layers and vertical continuity of nanopores are demonstrated using electron tomography. This work presents new routes for the construction of functional nanoporous media.

Multiple rereads of single proteins at single-amino acid resolution using nanopores

Henry Brinkerhoff et al.

Summary: This study presents a nanopore-based single-molecule peptide reader capable of identifying single amino acid substitutions within individual peptides with high accuracy. By pulling a DNA-peptide conjugate through a biological nanopore and reading the ion current signal, discrimination of single amino acid substitutions in single reads was achieved. The ability to obtain numerous independent reads of the same molecule and achieve a low error rate in amino acid variant identification signifies a promising advancement towards single-molecule protein fingerprinting and analysis technology.
