Dual Unlocking Strategy to Overcome Inefficient Nanomedicine Delivery and Tumor Hypoxia for Enhanced Photodynamic-Immunotherapy

Xianglong Li et al.

Summary: This study proposes a dual unlocking strategy to overcome the lack of blood vessels in solid tumors, using engineered hybrid nanoparticles and dexamethasone. The results show that pretreatment with dexamethasone can increase intratumoral micro-vessel density, enhance the delivery efficiency of the nanoparticles, and decrease HIF-1 alpha expression. Furthermore, the platinum in the nanoparticles can catalyze H2O2 to produce oxygen, addressing hypoxia and achieving efficient photodynamic therapy in vivo. Additionally, the nanoparticles can reverse the immunosuppressive tumor microenvironment and promote the therapeutic effect of anti-PD-L1 immunotherapy. This dual unlocking strategy provides an innovative approach to enhance nanomedicine delivery and hypoxia relief in tumor therapy.

GSH-Responsive Organosilica Hybrid Nanosystem as a Cascade Promoter for Enhanced Starvation and Chemodynamic Therapy

Huan Wu et al.

Summary: In this study, a glutathione-responsive organosilica hybrid micelle system was developed for enhanced starvation therapy and chemodynamic therapy. The system utilizes the catalytic activity of glucose oxidase to consume glucose and oxygen within tumor cells, generating hydrogen peroxide which is then decomposed to release oxygen. Simultaneously, the system can deplete glutathione in the tumor microenvironment and catalyze hydrogen peroxide to generate highly toxic hydroxyl radicals for chemodynamic therapy. The results demonstrate the significant antitumor efficacy of this system in both in vitro and in vivo experiments.

Nanocatalytic Theranostics with Glutathione Depletion and Enhanced Reactive Oxygen Species Generation for Efficient Cancer Therapy

Lian-Hua Fu et al.

Summary: Chemodynamic therapy (CDT) is a promising method for killing cancer cells by converting intracellular hydrogen peroxide (H2O2) into highly toxic hydroxyl radicals ((OH)-O-center dot). An intelligent nanocatalytic theranostics, PGC-DOX, was developed to address the limitations of endogenous H2O2 and high GSH levels in tumor cells, leading to efficient cancer therapy. The system combines H2O2 self-supply, GSH-elimination, and DOX-induced chemotherapy to effectively inhibit tumor growth with minimal side effects in vivo.

Caspase 3/GSDME-dependent pyroptosis contributes to chemotherapy drug-induced nephrotoxicity

Xiujin Shen et al.

Summary: This study investigated the impact of chemotherapeutic drugs cisplatin or doxorubicin on renal tubular epithelial cells, revealing that these drugs induce the cleavage of GSDME, leading to pyroptosis. By inhibiting GSDME, cell damage and inflammation caused by the drugs can be alleviated. The findings suggest that therapies targeting GSDME may be effective in reducing chemotherapeutic drug-induced nephrotoxicity.

Control of gasdermin D oligomerization and pyroptosis by the Ragulator-Rag-mTORC1 pathway

Charles L. Evavold et al.

Summary: The Ragulator-Rag complex is found to be necessary for GSDMD pore formation and pyroptosis in macrophages, promoting GSDMD oligomerization on the plasma membrane. Defects in GSDMD oligomerization and pore formation can be rescued by mitochondrial poisons that stimulate ROS production, and ROS modulation impacts the ability of inflammasome pathways to promote pore formation downstream of GSDMD cleavage.

Doxorubicin suppresses chondrocyte differentiation by stimulating ROS production

Cheng Wu et al.

Summary: DOX exacerbates OA progression and inhibits chondrocyte differentiation of BMMSCs, while also increasing ROS production. DOX further increases the elevation of plasma inflammatory factors induced by DMM, and significantly reduces the expression of chondrocyte differentiation-related markers.

Caspase-3-mediated GSDME induced Pyroptosis in breast cancer cells through the ROS/JNK signalling pathway

Ziwen Zhang et al.

Summary: The study investigated the potential link between DOX-induced cell death and pyroptosis in two human breast cancer cell lines (MDA-MB-231 and T47D), confirming that DOX treatment reduced cell viability and induced pyroptosis morphology. Protein expression analyses highlighted GSDME as a key regulator in DOX-induced pyroptosis and emphasized the role of Caspase-3 activation. Furthermore, DOX treatments induced intracellular accumulation of ROS, JNK phosphorylation, and Caspase-3 activation.
