Molecularly targeted nanomedicine enabled by inorganic nanoparticles for atherosclerosis diagnosis and treatment

Reactive oxygen species (ROS)-responsive size-reducible nanoassemblies for deeper atherosclerotic plaque penetration and enhanced macrophage-targeted drug delivery

Summary: Nanoparticle-based therapeutics show potential for treating atherosclerosis, but the complex plaque microenvironment hinders their targeting ability. In this study, we developed reactive oxygen species (ROS)-responsive and size-reducible nanoassemblies to overcome this barrier. These nanoassemblies specifically accumulated in atherosclerotic plaque, rapidly disassembled in response to excess ROS, and released smaller nanoparticles for deeper penetration and targeted drug delivery. In vivo studies demonstrated that the nanoassemblies reduced plaque size, lipid deposition, macrophage content, and local inflammatory factor levels, while also alleviating systemic inflammation. Overall, the ROS-responsive and size-reducible nanoassemblies exhibit enhanced targeting ability and provide an effective approach for atherosclerosis therapy.

Hyaluronic acid targeted and pH-responsive multifunctional nanoparticles for chemo-photothermal synergistic therapy of atherosclerosis

Summary: In this study, a multifunctional drug-loaded nano-delivery system with pH-responsive, CD44-targeted, and chemical-photothermal synergistic treatment was successfully prepared. This system showed excellent drug-controlled release properties in the weakly acidic microenvironment of atherosclerotic inflammation, and exhibited low cytotoxicity against RAW 264.7 cells. The combination of CuS photothermal treatment with anti-atherosclerosis chemotherapy had better effects than single treatment in vitro and in vivo, demonstrating broad application prospects in anti-atherosclerosis.

Summary: This study demonstrates that mild phototherapy, using foam cell-targeted nanoprobes, can effectively reduce intracellular lipid levels in foam cells without inducing apoptosis or necrosis. The protective effect against apoptosis or necrosis is correlated with the upregulation of heat shock protein 27. Additionally, the phototherapy regimen also inhibits intracellular cholesterol biosynthesis, excess cholesterol uptake, and triggers cholesterol efflux, ultimately preventing lipid accumulation in foam cells.
