Linking fluid-axons interactions to the macroscopic fluid transport properties of the brain

Mathematical Optimisation of Magnetic Nanoparticle Diffusion in the Brain White Matter

Summary: Magnetic nanoparticles (MNPs) can be used as a drug delivery system in brain diseases. However, achieving desired drug distribution patterns in the complex microstructure of brain tissues, particularly the white matter, is still unclear. In this study, a mathematical model was used to investigate the effects of key parameters on MNP diffusion in the brain white matter. The results show that a uniform distribution of MNPs can be achieved by adjusting particle size and magnetic field, and particle size plays a key role in determining MNPs' diffusion behaviors. Axon tortuosity may affect MNP transport, but this can be mitigated by applying an external magnetic field perpendicular to the local axon track.

Role of Tissue Hydraulic Permeability in Convection-Enhanced Delivery of Nanoparticle-Encapsulated Chemotherapy Drugs to Brain Tumour

Summary: Tissue hydraulic permeability can greatly affect drug accumulation and distribution in brain tumours. Mathematical modelling based on realistic tumour models extracted from patient images is used to predict drug delivery outcomes. Results show that infusing drugs into permeable tumours can improve drug transport and distribution, leading to more effective cell killing. Results also indicate that changes in tissue hydraulic permeability and blood pressure have a greater impact on drug delivery outcomes than fluid flow. Increasing fluid gain can enhance interstitial fluid flow and improve drug delivery outcomes. Different types of drugs show similar responses to changes in tissue hydraulic permeability.

On the microstructurally driven heterogeneous response of brain white matter to drug infusion pressure

Correlating the microstructural architecture and macrostructural behaviour of the brain

Summary: In this study, we investigated the mechanical behavior of brain tissue under different deformation modes, in different anatomical regions, and for varying axon orientations. The underlying microstructure of the tissue was characterized in terms of various constituents. We found that the shear modulus decreases with increasing myelin area fraction, the tensile shear modulus correlates positively with cell and neuronal area fraction, and the compressive shear modulus decreases with increasing glial cell area. Tissue non-linearity significantly depends on the myelin area fraction, cell density, and glial cell area.
