Radiation-Triggered Selenium-Engineered Mesoporous Silica Nanocapsules for RNAi Therapy in Radiotherapy-Resistant Glioblastoma

Summary: In this study, a biodegradable selenium-engineered mesoporous silica nano-capsule was developed for at-site RNA interference to inhibit the invasive growth of radiotherapy-resistant glioblastoma (rrGBM) and achieve maximum therapeutic benefit. The nanocapsule showed high stability, efficient blood-brain barrier penetration, and potent accumulation in rrGBM. It effectively reduced tumor growth and invasion, prolonging survival in an rrGBM model.

Pyruvate Facilitates FACT-Mediated γH2AX Loading to Chromatin and Promotes the Radiation Resistance of Glioblastoma

Summary: This study reveals a novel mechanism by which pyruvate produced by PKM2 enhances DNA repair by regulating gamma H2AX loading to chromatin, thus promoting the resistance of glioma cells to radiation therapy. PKM2 phosphorylation at S222 upon DNA damage and its interaction with the FACT complex play critical roles in this process. The levels of PKM2 S222 phosphorylation correlate with the malignancy and prognosis of human glioblastoma.

P300 promotes tumor recurrence by regulating radiation-induced conversion of glioma stem cells to vascular-like cells

Design of polymers for siRNA delivery: Recent progress and challenges

Summary: RNA interference technology has shown great potential in treating various diseases by specifically silencing target genes, but the primary challenge lies in efficiently delivering genetic materials such as siRNA to targeted tissues and cells in vivo. Recent research has focused on designing functional polymers to achieve effective siRNA delivery, addressing key parameters such as siRNA binding, serum stability, specific targeting, tissue penetration, cellular internalization, endosomal escape, and intracellular siRNA release. The beneficial roles of polymer nanostructures and hybrid nanocomposites in siRNA delivery have been reviewed, and challenges and future perspectives in polymer-based siRNA delivery systems have been discussed.
