Silk Microneedle Patch Capable of On-Demand Multidrug Delivery to the Brain for Glioblastoma Treatment

Zijing Wang et al.

Summary: A silk fibroin microneedle (SMN) patch designed for glioblastoma (GBM) can bypass the blood-brain barrier and release multiple drugs directly to the tumor site, providing an effective treatment strategy for brain tumors. The biocompatible and biodegradable patch allows for slow drug release over time and can be triggered remotely for rapid drug delivery. In animal models, the patch resulted in decreased tumor volume and increased survival rate, showing potential for clinical treatment of brain tumors.

PROTAC targeted protein degraders: the past is prologue

Miklos Bekes et al.

Summary: Targeted protein degradation (TPD) is a new therapeutic modality that can tackle disease-causing proteins which are difficult to target with conventional small molecules. PROTAC molecules, utilizing the ubiquitin-proteasome system to degrade target proteins, has achieved clinical proof-of-concept and attracted significant industry activity. Future directions include identifying target classes suitable for TPD, expanding the use of ubiquitin ligases for precision medicine, and extending the modality beyond oncology.

Bioinspired adhesive microneedle patch with gemcitabine encapsulation for pancreatic cancer treatment

Xiao Fu et al.

Summary: Inspired by the structures of octopus tentacles, researchers have developed a novel adhesive microneedle patch with gemcitabine encapsulation for the treatment of pancreatic cancer. The patch, made of gelatin methacryloyl, demonstrates good biocompatibility, flexibility, and drug loading efficiency. It can adhere to the irregular surfaces of tumors and efficiently release gemcitabine to penetrate tumor tissues, showing promising anti-tumor efficacy in vitro and in a mouse model.

E3 Ligase Ligands for PROTACs: How They Were Found and How to Discover New Ones

Tasuku Ishida et al.

Summary: PROTACs, or bifunctional degrader molecules, are a novel approach for understanding and treating human diseases by targeting specific proteins for degradation. Advances in nonpeptidic small-molecule E3 ligase ligands, such as VHL and CRBN, have revolutionized the field and led to the development of drug-like PROTACs with powerful degradation capabilities. Ongoing research aims to expand the chemical diversity of PROTACs and explore new approaches for targeted protein degradation.
