GSH-induced chemotaxis nanomotors for cancer treatment by ferroptosis strategy

Summary: In this study, a nanomotor based on a metal organic framework material was constructed by modifying polyethylene glycol and glutathione hydrolase on its surface. The nanomotor can control the level of glutathione in tumor cells, and it exhibits stable chemotaxis behavior in a complex environment. Loading the ferroptosis inducer enhances the induction effect of tumor ferroptosis. The nanomotor also shows enhanced delivery and penetration ability in tumors, enhancing the ferroptosis effect.

ACSL4 deficiency confers protection against ferroptosis-mediated acute kidney injury

Summary: This study explored the pathway mechanism of ferroptosis in acute kidney injury (AKI) by conducting RNA-sequence analysis. The results highlighted the important role of AcylCoA synthetase long-chain family (ACSL4) in promoting ferroptosis. Further investigations demonstrated that knockout of ACSL4 reduced ferroptosis and inhibited functional and pathological injury in AKI mice. Additionally, ACSL4 inhibition showed promising results in reducing AKI. This study provides novel insights into the pathogenesis of AKI and identifies ACSL4 as an effective therapeutic target.

Biodegradable Amorphous Copper Iron Tellurite Promoting the Utilization of Fenton-Like Ions for Efficient Synergistic Cancer Theranostics

Summary: In this study, a biodegradable a-CFT@IP6@BSA agent was developed for efficient cancer therapy. The agent can release plenty of Cu+ ions in a tumor-specific manner and utilize them for the production of hydroxyl radicals. It also depletes intracellular GSH to protect the hydroxyl radicals from scavenging. The treatment with a-CFT@IP6@BSA nanoparticles leads to the downregulation of glutathione peroxidase 4 and BCL-2, indicating the induction of ferroptosis and apoptotic death of cancer cells. The nanoparticles also demonstrate excellent anticancer efficiency in vivo and have the potential for imaging-guided cancer treatment.
