Clinical relevance of tumour-associated macrophages

Mikael J. Pittet et al.

Summary: The interaction of tumour-associated macrophages (TAMs) with cancer and stromal cells in the tumour microenvironment enables and sustains most of the hallmarks of cancer. The authors of this Review examine the diversity of TAMs in various cancer indications, which is being revisited with the advent of single-cell technologies, and discuss the functional roles of different TAM states, the prognostic and predictive value of TAM-related signatures as well as approaches involving TAMs that are currently being or will soon be tested in clinical trials.

Harnessing and Enhancing Macrophage Phagocytosis for Cancer Therapy

Siqi Chen et al.

Summary: Macrophage phagocytosis has emerged as a pivotal therapeutic strategy in cancer treatment. Targeting phagocytosis checkpoint blockade and using nanoparticles are effective ways to enhance macrophage phagocytosis, while chimeric antigen receptor macrophages serve as a next-generation therapeutic modality to induce efficacious anti-tumor immune responses by linking innate and adaptive immunity.

Fullerenols boosting the therapeutic effect of anti-CD47 antibody to trigger robust anti-tumor immunity by inducing calreticulin exposure

Kui Chen et al.

Summary: Fullerenol nanoparticles trigger CRT exposure by inducing ER stress in cancer cells, leading to efficient programmed cell removal. Combination therapy of fNP and anti-CD47 mAb shows significantly increased anti-tumor efficiency, even with half doses of anti-CD47 mAbs, indicating the potential role of fNP in reducing the amount of antibodies needed.

Macrophage-Mediated Tumor Cell Phagocytosis: Opportunity for Nanomedicine Intervention

Xuefei Zhou et al.

Summary: Macrophages, important in tumor immunity, can engulf tumor cells and present antigens; tumor cells evade macrophage phagocytosis through anti-phagocytic molecules, while macrophages' efferocytosis of apoptotic cells promotes tumor immune escape; blocking anti-phagocytic signals or inhibiting tumor efferocytosis is a potential strategy for cancer immunotherapy.
