The development of inhibitors of leucine-rich repeat kinase 2 (LRRK2) as a therapeutic strategy for Parkinson's disease: the current state of play

Sonia Azeggagh et al.

Summary: Current therapeutic approaches for Parkinson's disease focus on symptom relief rather than disease progression. Inhibition of LRRK2 function is a promising strategy due to its role in familial and idiopathic Parkinson's disease. Progress has been made in finding effective LRRK2 inhibitors, but the complexity of LRRK2 presents challenges and opportunities for unintended consequences. Multiple molecules are in clinical trials, providing cause for optimism in the search for alternative treatment options.

Targeting the WASF3 complex to suppress metastasis

Ameya J. Limaye et al.

Summary: WASF proteins regulate the dynamics of the actin cytoskeleton, playing a crucial role in cancer metastasis and invasion. The WASF3 regulatory complex correlates with metastatic potential and survival rate, suggesting that targeting the complex may be a novel strategy for suppressing metastasis.

Preclinical and clinical evaluation of the LRRK2 inhibitor DNL201 for Parkinson's disease

Danna Jennings et al.

Summary: Mutations in LRRK2 are common genetic risk factors for Parkinson's disease. Inhibition of LRRK2 with DNL201 has shown positive effects in preclinical and clinical trials, improving lysosomal function and demonstrating safety and tolerability. Further development of LRRK2 inhibitors as a therapeutic modality for Parkinson's disease is warranted.

Allosteric Inhibition of Parkinson's-Linked LRRK2 by Constrained Peptides

Leah G. Helton et al.

Summary: Dimerization of LRRK2 can be inhibited by specific constrained peptides, leading to decreased kinase activity, reduced ROS production, and inhibition of PD-related apoptosis in primary cortical neurons. Unlike other inhibitors, these peptides do not affect the cellular localization of LRRK2.

Structural analysis of the full-length human LRRK2

Alexander Myasnikov et al.

Summary: Mutations in LRRK2 are commonly associated with Parkinson's disease. This study provides insights into the physiological and pathological roles of LRRK2, and establishes a structural template for future therapeutic interventions in PD. The high-resolution structures of full-length human LRRK2 and COR-mediated LRRK2 dimers reveal key elements for rationalizing disease-causing mutations and potential targets for inhibitors.
